三拗汤调节溃疡性结肠炎模型大鼠肺与结肠组织细胞膜AQP1、AQP4的表达效应 被引量:5

摘要 目的:观察三拗汤对溃疡性结肠炎(ulcerative colitis,UC)大鼠肺与结肠组织细胞膜AQP1、AQP4的表达效应。方法:将60只SD大鼠按照随机数字表法分为空白组、模型组和给药组,每组各20只。采用TNBS+乙醇法7周制备溃疡性结肠炎大鼠模型,检测各组大鼠粪水含量、肺功能及血气指标;免疫组织化学法、Western blot法检测大鼠结肠组织AQP4与肺组织AQP1蛋白表达。结果:模型组粪水含量高于空白组,给药组粪水含量低于模型组,差异均有统计学意义(P<0.05)。模型组PCO2高于空白组,PH值、PO2低于空白组;给药组PO2、PH值高于模型组,PCO2低于模型组,差异均有统计学意义(P<0.05)。模型组气道阻力及肺顺应性高于空白组,给药组降低气道阻力及肺顺应性低于模型组,差异均有统计学意义(P<0.05)。模型组结肠组织AQP4蛋白半定量及肺组织AQP1表达低于空白组及给药组,差异具有统计学意义(P<0.05)。结论:三拗汤治疗溃疡性结肠炎模型大鼠,肺组织AQP1表达上升,促进肺泡水清除,减轻肺损伤,改善呼吸功能;同时结肠组织AQP4表达明显上升,粪水含量降低,泄泻减轻,共同降低大鼠死亡率。 Objective: To observe the influence of Rough and Ready Three Decoction on the expression of AQP1 and AQP4 in lung and colon tissue membranes of ulcerative colitis(UC) model rats. Methods: Sixty SD rats were randomly divided into the blank group, the model group and the administration group, with 20 rats in each group.The rat model of UC was established by TNBS + ethanol for 7 weeks. The fecal water content, pulmonary function and blood gas were measured in each group. The expression of AQP4 and AQP1 in colon tissue and lung tissue were detected by immunohistochemistry and Western blot. Results: The fecal water content of model group was higher than that of the blank group, and that of administration group was lower than that of the model group, and all the differences were statistically significant(P<0.05). PCO2 in the model group was higher than that in the blank group, PH and PO2 were lower than those in the blank group, PO2 and PH in the administration group were higher than those in the model group, PCO2 was lower than that in the model group, and all the differences were statistically significant(P<0.05). The airway resistance and pulmonary compliance of the model group were higher than those of the blank group, while those of the administration group were lower than those of the model group, and all the differences were statistically significant(P<0.05). Semi-quantitative expression of AQP4 protein in colon tissue and expression of AQP1 in lung tissue of the model group were lower than those of the blank group and the administration group, and all the differences were statistically significant(P<0.05). Conclusion: Rough and Ready Three Decoction has a remarkable efficacy on UC, can increase the expression of AQP1 in lung tissue, promote alveolar water clearance, alleviate lung injury and improve respiratory function in UC model rats;at the same time, it can significantly increase the expression of AQP4 in colon tissue, reduce the content of fecal water, relieve diarrhea and reduce the mortality rate of rats.
