摘要
目的观察白细胞介素(IL)-29、Toll样受体(TLR)4在过敏性紫癜(HSP)患儿中的表达及临床意义。方法选择48例HSP患儿作为观察组,20例健康儿童作为对照组,实时荧光定量PCR检测对照组和观察组(急性期、恢复期)外周血单个核细胞(PBMC)中IL-29mRNA、TLR4mRNA的表达水平,并检测不同时期血清中IL-29、TLR4、IL-4、免疫球蛋白(Ig)A、IgM、IgG、C3、C4、β2微球蛋白(β2-MG)水平。结果观察组急性期PBMC中IL-29、TLR4mRNA相对表达水平,以及血清中IL-29、TLR4水平明显高于恢复期和对照组,差异均有统计学意义(P<0.05)。观察组急性期IgA、C3、IL-4、β2-MG水平明显高于对照组,差异均有统计学意义(P<0.05)。血清中IL-29与TLR4呈显著正相关(P<0.05);血清中IL-29与IgA、C3、IL-4、β2-MG呈显著正相关(P<0.05);血清中TLR4与IgA、C3、IL-4、β2-MG呈显著正相关(P<0.05)。结论IL-29、TLR4参与了HSP的发生、发展,维持了机体的炎症水平。
Objective To observe the expression and clinical significance of interleukin(IL)-29 and Toll-like receptor(TLR)4 in children with allergic purpura.Methods A total of 48 children with allergic purpura were selected as observation group,20 healthy children were recruited as control group,FQ-PCR was used to detect IL-29 mRNA and TLR4 mRNA in peripheral blood mononuclear cell(PBMC)of control group and observation group(different stages including acute phase,recovery phase).The expression of IL-29,TLR4,IL-4,IgA,IgM,IgG,C3,C4,β2-microglobulin(β2-MG)in serum were detected at different times.Results The relative expression levels of IL-29 mRNA and TLR4 mRNA in the PBMC of the observation group were significantly higher than those in the recovery period of observation group and the control group.The levels of IL-29 and TLR4 in serum were significantly higher than those in the recovery period of observation group and the control group(P<0.05).The levels of IgA,C3,IL-4 andβ2-MG in the observation group were significantly higher than those in the control group(P<0.05).Serum IL-29 and TLR4 had positive correlation(P<0.05).Serum IL-29 positively correlated with IgA,C3,IL-4,β2-MG(P<0.05).Serum TLR4 positively correlated with IgA,C3,IL-4,β2-MG(P<0.05).Conclusion IL-29 and TLR4 are involved in the occurrence and progression of allergic purpura and maintain the body′s inflammation level.
