摘要 以‘凤丹’牡丹种胚为试验材料,研究不同的外植体处理时间、激素配比、基本培养基、光照强度及不同成熟度的种胚对‘凤丹’牡丹胚培养的影响,并探究愈伤组织诱导过程中最佳的防褐方式。结果表明:‘凤丹’牡丹无菌体系建立过程中,种子用0.1%HgCl消毒8min时,其萌芽率最高,污染率、褐化率最低,为最佳消毒时间。胚培养过程中,最佳激素配比为0.5mg/L6-BA+0.1或0.2mg/LNAA;并以MS为基本培养基,光照强度为1000Lux时最适合种胚的生长;且以6月底采摘的幼嫩种子的种胚为试验材料时,萌芽率最高,污染率最低,可作为胚培养过程中的理想试材。防褐研究结果表明,AC与PVP均在不同程度上降低了愈伤组织的褐化率,其中PVP在2g/L的水平上愈伤组织出愈率最高,褐化率最低。 The embryos of Paeonia ostii ‘Feng Dan’ were used aSexperimental materialSto study the effects of different explant treatment time,hormone ratio,basic culture media,light intensity and seeds with different maturity on embryo culture of Paeonia ostii ‘Feng Dan’,and try to find the best way to prevent browning in callus induction.The results showed that during the establishment of aseptic system,when seeds were sterilized with 0.1%HgCl for 8min,the germination rate was the highest,the contamination rate and browning rate were the lowest.In the process of embryo culture,the best ratio of hormone was 0.5mg/L of 6-BA and 0.1mg/L or 0.2mg/L of NAA.MSbasic medium and the light intensity of1 000lux was the most suitable for the growth of embryo.Using the young embryo picked at the end of June,the germination rate was the highest and the contamination rate was the lowest.It can be used aSan ideal material for embryo culture.In terms of the best way to prevent browning during calluSinduction,this study indicated that both AC and PVP reduced the browning rate of callus.When2g/L PVP was added,the introducement rate of callus was the highest and the browning rate was the lowest.
