摘要
叶色突变是植物最常见的突变性状,其形成机理多样。前期Cinnamoyl-coAReductase1(BpCCR1)基因的过表达白桦转化株系中发现一个黄叶突变株系(yl),该株系全株叶片颜色在整个生长季均呈现黄色。试验以黄叶突变株yl、野生型白桦WT和转BpCCR1基因OE11株系为材料,测定叶片光合色素含量,叶色参数和光合参数,并采用转录组测序对差异LncRNA及其靶基因进行分析。结果表明:与对照株系(WT和OE11)相比,yl株系的叶绿素a、叶绿素b、总叶绿素含量、类胡萝卜素含量和净光合速率降低,而yl株系的叶色参数的绝对值升高。此外,与WT相比,132个LncRNA在yl株系中差异表达。与WT相比,137个LncRNA在OE11株系中差异表达。与OE11相比,160个LncRNA在yl株系中差异表达。这些差异表达的LncRNA调控大量的靶基因并参与到卟啉和叶绿素生物代谢、类胡萝卜素生物合成及光合作用等代谢通路。因此,这些差异LncRNA及其靶基因可能与黄叶突变株叶色变异的形成密切相关。
Leaf color mutation is the most common mutant trait in plants, and the molecular mechanisms underlying the variability of leaf color are extremely diverse. Our previous study has revealed a yellow leaf mutant line (yl) among the overexpression lines of Cinnamoyl-coA Reductase1 (CCR1) gene in birch, with yellow color in the whole plant body throughout the growing season. In our work, plants of yellow leaf mutant line (yl), wild type (WT) and transgenic BpCCR1 gene OE11 line were used for materials to determine leaf photosynthetic pigment content, leaf color parameters, photosynthetic parameters, and analyze differential LncRNA and their target genes using transcriptome sequencing. The results showed that compared to the control lines (WT and OE11), the chlorophyll a, chlorophyll b, total chlorophyll content, carotenoid content and net photosynthetic rate of yl line were decreased, while the absolute value of leaf color parameters of yl line was increased. Contrary to WT, 132 LncRNAs were differentially expressed in yl line. Compared with WT, 137 LncRNAs were differentially expressed in OE11 line. Compared with OE11, 160 LncRNAs were differentially expressed in yl line. Meanwhile, these LncRNAs expressed differentially could regulate a large number of target genes and were involved in many metabolic pathways such as porphyrin and chlorophyll metabolism, carotenoid biosynthesis and photosynthesis. Therefore, these LncRNAs and their target genes identified from our transcriptome profiles might be associated with leaf color variation in birch.
