摘要
目的研究丹参对电离辐射致SD大鼠损伤的防护机制。方法50只SD大鼠随机分为正常组、辐射组(辐射模型)、丹参低剂量组(125mg/kg)、中(250mg/kg)、高剂量(375mg/kg)治疗组各10只。除正常对照组外,其余各组采用小剂量多次照射(每次照射10CGy,间隔24h连续照射5次),累积辐射剂量为0.5Gy。丹参治疗各组在照射当天给予药物灌胃,正常对照组和辐射模型组灌等量生理盐水,每日1次,持续20d,第20d检测外周血象和脑组织丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽(GSH)。结果与正常对照组比较,辐射组外周血象值和SOD、GSH活性减低,MDA含量升高,差异有统计学意义(P<0.01);与模型组比较,丹参治疗组外周血象值和SOD、GSH活性增加,MDA含量减低(bP<0.05或cP<0.01)。结论丹参片对低剂量电离辐射损伤有一定的防护作用,其机制可能与丹参改善自由基代谢,提高机体抗氧化损伤能力有关,为辐射防护的中药方案提供了数据支撑。
Objective To study the protective mechanism of Danshen Tablets on SD rats injured by ionizing radiation. Methods Fifty SD rats were randomly divided into normal group, radiation group (radiation model), Salvia miltiorrhiza low dose group (125 mg/kg), medium dose group (250 mg/kg) and high dose group (375 mg/kg) with 10 rats in each group. Except for the normal control group, the other groups were treated with low dose multiple irradiation (10 CGy each time, 5 irradiations at 24-hour intervals), and the cumulative radiation dose was 0.5 Gy. Salvia miltiorrhiza treatment groups were given medication on the day of irradiation. Normal control group and radiation model group were given the same amount of saline once a day for 20 days. Malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione (GSH) in peripheral blood and brain tissue were detected on the 20th day. Results Compared with the normal control group, the peripheral blood image value and SOD, GSH activity decreased, and MDA content increased in the radiation group (P < 0.01). Compared with the model group, the peripheral blood image value, SOD and GSH activity increased and MDA content decreased in the salvia miltiorrhiza treatment group (bP < 0.05 or cP < 0.01). Conclusion Salvia miltiorrhiza tablets have protective effect on low dose ionizing radiation damage. Its mechanism may be related to the improvement of free radical metabolism and the enhancement of anti-oxidative damage ability of organism by Salvia miltiorrhiza. It provides data support for the traditional Chinese medicine scheme of radiation protection.
