高效液相色谱法测定百癣夏塔热片浸膏中没食子酸的含量 被引量:2

Determination of gallic acid content of Baixuan Xiatare Tablet extract by high performance liquid chromatography

摘要 目的建立用高效液相色谱法测定百癣夏塔热片浸膏中没食子酸含量的方法,以便更好地控制百癣夏塔热片的质量。方法,以没食子酸为标准品,将百癣浸膏加甲醇超声提取后,采用色谱条件:色谱柱为EclipseplusCig(4.6mmX250mm,5μm),流动相为甲醇-0.1%磷酸(6:94),流速为1.000ml/min,检测波长为270nm,柱温为30℃,进样量为10μl。结果在该色谱条件下,没食子酸在10. 55 μg/ml ~ 84.40 μg/ml浓度内呈良好的线性关系,r=0.9996,高、中、低3个质量浓度的加样回收率平均为99.72%,RSD为1.64%,仪器精密度良好,供试品溶液稳定性良好,在室温下24h内稳定,该方法重复性较好。结论该方法色谱峰之间能有效的分离,精密度、稳定性、重复性较好,回收率符合规定,可用于百癣夏塔热片浸膏没食子酸含量的测定和质量控制。 Objective To establish a method for determination of gallic acid in the extract of Baixuan Xiatare Tablets by high performance liquid chromatography ( HPLC), so as to ensure the quality of Baixuan Xiatare Tablets. Methods Gallic acid was used as standard substance, and the Baixuuan extract was extracted by ultrasound with methanol. The chromatographie condi-tions were as followings: the chromatographic column was Eclipse plus C18 (4.6 mm x250 mm, 5μm), the mobile phase was methanol-0.1% phosphoric acid (6 : 94),the flow rate was 1, 000 ml/ min, the detection wavelength was 270 nm, the column temperature was 30℃, and the injection volume was 10 μl. Results Under this chromatographic condition, gallic acid showed a good linear relationship in the concentration range of 10. 55 μg/ ml -84. 40 μg/ml, r=0. 9996. The average recovery of high, medium and low concentration was 99. 72%,RSD= 1.64%. The instrument has good precision, good stability of the, sample solution and stability within 24 hours at room temperature. The method has good repeatability. Conclusion The method can be used for the determination and quality control of gallic acid in Baixuan Xiatare tablet extract.
