天麻素对小鼠放射性心脏损伤的保护作用 被引量:2

摘要 目的探讨天麻素对小鼠放射性心脏损伤的保护作用。方法50只C57BL/6小鼠随机分为对照组,单纯照射组,照射+天麻素低、中、高剂量干预组(50、100、150mg/kg)。各实验组小鼠利用放疗加速器造模(照射剂量18Gy)。药物干预组小鼠于照射前三天开始每天于腹腔注射天麻素注射液一次。于照射后4周末,用ELISA法检测血清IL-6,TUNEL法检测心肌细胞凋亡率,Westernblot法检测心肌组织caspase-3水平,比色法检测超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果与对照组比较,单纯照射组小鼠心肌细胞凋亡率和IL-6、caspase-3、MDA表达水平明显升高,SOD表达水平明显减弱,差异具有统计学意义(P<0.05);与单纯照射组比较,中、高剂量药物干预组心肌细胞凋亡率和IL-6、caspase-3及MDA的表达水平明显减弱,SOD表达水平明显增强,差异具有统计学意义(P<0.05);与单纯照射组比较,低剂量药物干预组IL-6的表达减弱,具有统计学意义(P<0.05),心肌细胞凋亡率和caspase-3及SOD、MDA的表达水平差异无统计学意义(P>0.05)。结论天麻素可减轻小鼠心肌细胞的放射性心脏损伤:可能通过下调心肌细胞氧化应激反应程度、降低细胞凋亡及抗炎来实现保护作用。 Objective To investigate the protective effect of Gastrodin on radiation-induced heart injury in mice.Methods 50 C57BL/6 mice were randomly divided into control group,simple irradiation group,irradiation+gastrodin low,medium and high dose intervention group(50,100,150mg/kg).Mice in each experimental group were modeled using a radiotherapy accelerator(radiation dose 18Gy).Mice in the drug intervention group were intraperitoneally injected with gastrodin injection once a day starting three days before irradiation.At the end of 4 weeks after irradiation,serum Interleukin-6(IL-6)was detected by enzyme linked immunosorbent assay(ELISA).Transferase-mediated deoxyuridine triphosphate-biotin nick end labeling(TUNEL)assay was used to detect the apoptosis rate of myocardial cells.Western blot was used to detect the expression of caspase-3;Superoxide dismutase(SOD)and malondialdehyde(MDA)were detected by colorimetric method.Results Compared with the control group,the apoptosis rate,IL-6,caspase-3 and MDA expression levels of myocardial cells were significantly increased and SOD expression was significantly decreased in the single irradiation group,showing statistical significance(P<0.05).Compared with the single irradiation group,the apoptosis rate,IL-6,caspase-3 and MDA expression were significantly decreased and SOD expression was significantly increased in the medium-dose and high-dose gastrodin intervention groups,with statistically significant differences(P<0.05).Compared with the simple irradiation group,the expression of IL-6 in the low-dose drug intervention group was decreased,which was statistically significant(P<0.05),and there was no statistically significant difference in the apoptosis rate of cardiac myocytes,caspase-3,SOD and MDA expression levels(P>0.05).Conclusion Gastrodin can alleviate the radiation heart injury of mouse cardiac myocytes,and may achieve the protective effect by down-regulating the oxidative stress response of cardiac myocytes,alleviating apoptosis and anti-inflammation.
