减压-超声波联用提取红松籽油动力学模型及油脂组成、抗氧化活性研究 被引量:7

摘要 为探究减压-超声联用技术对红松籽油提取效果的影响,并考察提取出的红松籽油的抗氧化活性,本研究以红松籽油得率为指标,在减压-超声波联用提取红松籽油单因素试验的基础上,重点探讨了不同超声波功率在不同时间条件下对红松籽油提取效果的影响,对提取过程进行模型拟合并验证。测定减压-超声波联用提取的红松籽油的脂肪酸组成及含量,并测定其清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)和2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(2, 2’-azino-bis-3-ethylbenzothiazoline-6-sulfonnic acid,ABTS)自由基的能力。研究结果表明以正己烷为最适提取溶剂,在真空度0.08~0.09 MPa条件下,超声波提取红松籽油的最佳工艺参数为:超声波功率210 W、料液比1:7(g:mL)、超声温度45℃、超声处理时间25 min,此条件下松籽油得率为(53.01%±0.30%)。试验选取的四种动力学模型中,玻尔兹曼模型更好的拟合减压-超声波联用提取动力学过程(R^2≥0.9798)。提取的红松籽油含有46.27%的亚油酸,28.95%的油酸以及13.26%的皮诺敛酸。抗氧化试验结果表明:红松籽油对DPPH自由基和ABTS+自由基清除能力的IC50值分别为(8.20±0.77)mg/mL、(7.74±0.06)mg/mL。减压-超声波联用通过降低容器内压强,减少溶液间分子压力,进而降低提取过程中的能耗,缩短提取时间,降低提取温度,有利于保护热敏性物质的抗氧化活性。 In order to explore the effect of decompression assisted ultrasonic extraction on the Korean pine seed oil, and to investigate the antioxidant activity of the extracted Korean pine seed oil, the study used the yield of Korean pine seed oil as an indicator. Based on the single factor experiment of extracting Korean pine seed oil, the effects of different ultrasonic power on the extraction of Korean pine seed oil under different time conditions were discussed. The extraction process was modeled and verified The fatty acid composition was measured and evaluate 1, 1-dipheny l-2-picrylhydrazyl(DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid(ABTS)of Korean pine seed oil extracted by decompression assisted ultrasonic. The results for ultrasonic extraction under the vacuum degree of 0.08-0.09 MPa were as follows: the best extraction solvent was n-hexane,solid-liquid ratio was 1:7(g:mL), ultrasonic power was 210 W, the four kinetic models selected in the experiment, the Boltzman model can better fit the dynamic process of Korean pine seed oil extracted by decompression assisted ultrasonic(R^2≥0.9798). The Korean pine seed oil extracted by decompression assisted ultrasonic contained linoleic acid was 46.27% oleic acid was 28.95% and pinolenic acid was 13.26%. The results of antioxidant tests showed that: the IC50 for DPPH free radicals and ABTS+ radicals were respectively(8.20±0.77) mg/mL and(7.74±0.06) mg/mL. The Korean pine seed oil extracted by decompression assisted ultrasonic had a certain antioxidant activity. The decompression assisted ultrasonic reduces the pressure in the container and reduces the molecular pressure between the solutions, thereby reducing the energy consumption during the extraction process, shortening the extraction time, and lowering the extraction temperature which protects the antioxidant activity of the heat sensitive substance.
