摘要
采用高效液相色谱(HPLC)-示差折光检测法和斐林试剂滴定法检测绵白糖中还原糖分的含量,分析比较二者的测定结果及可操作性。结果表明,HPLC-示差折光检测法可操作强,与斐林试剂滴定法相关系数为0.983(P<0.05),具有相关性显著;斐林试剂滴定法平行样检测RSD值为0.11%~3.1%,而HPLC-示差折光检测法平行样检测RSD值为0.06%~0.9%。斐林试剂滴定法易受干扰因素如试剂溶液颜色的影响较大,终点不易判定,而HPLC-示差折光检测法操作简单、快速,受干扰因素影响相对较小。本研究为HPLC-示差折光法的准确性提供数据论证,有助于推动HPLC-示差折光法在快速检测绵白糖还原糖分的应用,提高制糖企业日常检验分析的效率。
The analysis of the reducing sugar content in soft sugar were carried out by the methods of HPLC-refractive index detection and the Fehling reagent titration, respectively, the results and maneuverability of two methods were compared. The results showed that HPLC-refractive index detection method could be operated strongly, the correlation coefficient was 0.983 (P<0.05) between these two methods with a statistically significant correlation. The RSD value of the parallel analysis for HPLC-refractive index detection method was 0.06%~0.9%, in comparison with the value of 0.11%~3.1% by Fehling reagent titration method. According to the results, it was more sensitive to the disturbing factors such as reagent solution’s color, and the end of reaction was difficult to judge using Fehling reagent titration method. Easy operation and a fast reading with a smaller deviation for the results made it more feasible for the analysis by HPLC-refractive index detection method. The results evaluated that the reliability on the analysis for the contents of reductive sugar in soft sugar by HPLC-refractive index detection method, which promoted the efficiency of regular inspection in sugar manufactories.
