脑脉利颗粒对斑马鱼血管新生的促进及对血小板聚集性血栓形成的预防作用研究 被引量:10

摘要 目的 初步研究脑脉利颗粒对斑马鱼血管新生的促进作用与机制,及其对血小板聚集性血栓形成的预防作用,为临床用药提供更有价值的理论支持。方法 通过对斑马鱼肠下血管面积、肠下血管出芽数、总RNA浓度和纯度、心脏红细胞染色强度等指标,评价脑脉利颗粒对斑马鱼血管新生的促进作用和血小板聚集性血栓形成的预防作用。结果 脑脉利颗粒在250和500μg/ml浓度时有显著的血管新生促进作用,且在500μg/ml浓度时能明显上调VEGFR1基因表达(P<0.01),在1000μg/ml浓度时有显著的血栓形成预防作用(P<0.01)。结论 脑脉利颗粒具有显著的血管新生促进作用,机制可能与上调VEGFR1的表达有关,且对血栓形成有显著的预防作用。 Objective To provide more valuable evidence for clinical medication, the mechanism of promoting angiogenesis and preventing platelet aggregation thrombosis of NaoMaiLi are studied. Methods The promotion and the preventability of NaoMaiLi Granule on angiogenesis and platelet aggregation thrombosis respectively in zebrafish are evaluated through the indexes of subintestinal vascular area and number of buds, the relative expression of VEGFR1 gene, and the staining strength of cardiac red blood cells. Results NaoMaiLi Granule has a significant promoting effect at concentrations of 250 and 500 μg/ml, and VEGFR1 gene expression is significantly increased at concentrations of 500 μg/ml(P < 0.01). Meanwhile the drug also has a preventability at concentration of 1 000 μg/ml (P < 0.01). Conclusion NaoMaiLi Granule have a significant role in promoting angiogenesis, which may be related to the up-regulation of VEGFR1 expression, meanwhile it has a significant preventability on platelet aggregation thrombosis.
