摘要
采用带正电的表面活性剂十二烷基三甲基溴化铵(DTAB)与带负电的Hg(Ⅱ)核酸适配体(5′-TTCTTTCTTCCCTTGTTTGTT-3′),通过静电作用与疏水作用结合为超分子组装体,在组装体中包埋荧光染料尼罗红作为荧光探针。当加入待测物Hg(Ⅱ)后,Hg(Ⅱ)与胸腺嘧啶T形成"T-Hg(Ⅱ)-T"发夹状结构,从而诱导其解组装。利用尼罗红在水环境与疏水腔中荧光强度的不同构建"荧光关"的Hg(Ⅱ)的检测体系。该检测体系对Hg(Ⅱ)的检出限达到5.1×10-12 mol·L-1,线性检测范围为1.0×10-11~1.0×10-10 mol·L-1。其它离子对检测体系的影响可基本忽略不计,检测体系具有良好的选择性。方法可用于自来水中Hg(Ⅱ)的检测,回收率为92%~107%。
Driven by electrostatic and hydrophobic interaction,positively charged surfactant N,N,N-trimethyl-1-dodecanaminium bromide(DTAB) and negatively charged Hg(Ⅱ) aptamer(5′-TTCTTTCTTCCCTTGTTTGTT-3′) would form micellar-type assemblies.The hydrophobic dye nile red was encapsulated in the apolar interiors of the supramolecular assemblies as a fluorescent probe.Upon addition of target Hg(Ⅱ),the Hg(Ⅱ) aptamer with thymine(T)-rich sequences selectively bound with Hg(Ⅱ) to form an Hg(Ⅱ)-bridged T base pair and the aptamer changed into a hairpin-like structure.The fluorescence of nile red was turned off due to the disassembling of the supramolecular assemblies.The detection limit of Hg(Ⅱ) was as low as 5.1×10-12 mol·L-1 with a good selectivity,and the linear detection range was from 1.0×10-11 to 1.0×10-10 mol·L-1.This method revealed good recovery rates from 92% to 107% in tap water.
