经SDF-1转染的间充质干细胞对高糖所致足细胞损伤的修复作用 被引量:2

摘要 目的体外研究经基质细胞衍生因子(SDF)-1转染的间充质干细胞(MSCs)是否更容易向损伤足细胞迁移并修复损伤足细胞。方法高糖诱导建立肾小球足细胞损伤模型。MSCs与肾小球足细胞用Transwell小室共培养,做迁移实验,检测SDF-1转染的MSCs是否更容易迁移至损伤足细胞侧。并检测足细胞中胰岛素样生长因子(IGF)-1、转化生长因子(TGF)-β、半胱氨酸-天冬氨酸蛋白酶(caspase)-3的表达水平。结果经SDF-1转染的MSCs更易迁移至受损足细胞侧,且可下调受损足细胞中IGF-1、TGF-β、caspase-3表达。结论经SDF-1转染的MSCs能够更容易迁移到受损的足细胞侧,且对高糖所致足细胞损伤有修复作用。 Objective To investigate the repair effects of mesenchymal stem cells transfected with SDF-1 on podocyte injury induced by high glucose. Methods The glomerular podocyte injury model was induced by hyperglycemia.MSCs and glomerular podocytes were co-cultured in transwell Chambers for migration experiments to detect whether sdf-1-transfected MSCs were more likely to migrate to the podocyte injured side.The expression levels of IGF-1,TGF-1 and caspase-3 were detected. Results SDF-1-transfected MSCs were more likely to migrate to the injured podocytes,and down-regulated the expressions of IGF-1,TGF-β and caspase-3 in the injured podocytes. Conclusions SDF-1-transfected MSCs could more easily migrate to the damaged podocyte side and repair the damage caused by high glucose.
