摘要
本试验针对当前对养猪业危害严重的猪瘟病毒(CSFV)、猪繁殖与呼吸综合征病毒(PRRSV)、猪圆环病毒2型(PCV2)、伪狂犬病病毒(PRV)和猪细小病毒(PPV),分别建立了检测CSFV和PRRSV的双重RT-PCR方法和检测PCV2、 PRV及PPV的多重PCR方法。所建立的方法对 CSFV、PRRSV、PCV2、PRV和 PPV核酸的检出量分别为 7.5 pg/Μl、14.2 pg/Μl、6.9 pg/Μl、9.2 pg/Μl 和 8.6 pg/Μl。应用建立的方法分别对陕西省的陕南、陕北、关中等地区部分猪场采集的118份血清进行病毒检测。5种病毒的检出率分别是,CSFV为 4.23%(5/118),PRRSV为 22.01%(26/118),PCV2 为15.25%(18/118),PRV为 22.88%(27/118),从血清样品中未检测到 PPV。本研究建立的检测RNA 病毒双重RT-PCR和Dna 病毒的多重PCR方法,为猪主要病毒其快速检测提供了可选择方法。
This study aims to develop the duplex RT-PCR method for detecting classical swine fever virus(CSFV)and porcine reproductive and respiratory syndrome virus(PRRSV) as well as multiplex PCR method for porcine circovirus type 2(PCV2), pseudorabies virus(PRV) and pig parvovirus(PPV), which can cause enormous economic losses. The established methods for the detection of CSFV, PRRSV, PCV2, PRV and PPV nucleic acids were 7.5 pg/μL, 14.2 pg/μL, 6.9 pg/μL, 9.2 pg/μL and 8.6 pg/μL, respectively.We then tested 118 serum samples collected from some pig farms in Southern Shaanxi, northern Shaanxi and Guanzhong regions by using the developed methods and the results showed that the detection rates of the five viruses were 4.23%(5/118) for CSFV, 22.01%(26/118) for PRRSV, 15.25%(18/118) for PCV2, and 22.88% for PRV(27/118), respectively. However, the PPV was not detected. The established multiplex PCR method for detecting DNA viruses while duplex RT-PCR for RNA viruses can be used to detect the main pathogenic virus of swine and provide another option for rapid detection.
