摘要
为JY24菌株的中试生产和产品研发提供理论依据,采用单因素试验与正交试验相结合的方法筛选JY24菌株摇瓶培养条件和培养基成分,并在此基础上优化培养基成分。结果表明:JY24菌株发酵条件为转速240r/min,温度35℃,初始pH7.0,装液量10%,接种量3%,发酵时间36h;最佳培养基组成为蔗糖12.0g/L+酵母膏12.0g/L+磷酸二氢钾1.25g/L+氯化钙0.22g/L+硫酸镁0.30g/L。在此条件下,发酵液活菌数达1.73×10^10CFU/mL。
To provide theory basis for middle trial and product research and development of JY24 strain,shake flask culture conditions and composition of culture medium of feed Bacillus subtilis JY24 strain were studied and optimized by single factor methods combining orthogonal design,afterwards,the composition of culture medium was optimized. Results: The culture conditions included rotation speed 240 r/min ,temperature 35℃,initial pH 7.0,liquid volume 10%,inoculation amount 3%,fermentation time 36 h. And the optimal compositions of medium were sucrose 12.0 g/L +yeast extract 12.0 g/L+KH 2 PO 4 1.25 g/L+CaCl 2 0.22 g/L+MgSO 4 ·7H 2 O 0.30 g/L. Under these conditions,the living bacteria amount can reach 1.73×10^10 CFU/mL.
