摘要
目的 :观察左旋咪唑对实验性变态反应性脑脊髓炎 (EAE)中枢神经系统星形胶质细胞的影响。方法 :用豚鼠全脊髓匀浆免疫Wistar大鼠建立EAE动物模型 ,应用免疫组化法观察左旋咪唑在不同给药方式(1次给药或 3次给药 )和不同剂量 (分别为 5、1 0、30mg/kg)的情况下对EAE大鼠炎性脱髓鞘病灶内或周围星形胶质细胞数量和形态影响的变化。结果 :左旋咪唑可提高Wistar大鼠EAE的发病率 ,加重其临床症状 ,在免疫后 1 6d ,炎性脱髓鞘病灶内和周围星形胶质细胞数量增多 ,胞体肥大 ,其中 3次给药的作用强于 1次给药 ,在 3次给药时剂量为 1 0mg/kg时最重。结论
Objective: To observe the effects of levamisole on astrocytes of the central nervous system (CNS) in experimental allergic encephalomyelitis(EAE). Methods:EAE in Wistar rats immunized with spinal cords homogenate of gui nea pigs(GPSCH) and complete Freund's adjuvant(CFA) was induced.EAE induced rats received levamisole intraperitoneally on day 0 post immunization or days 0,1 ,2 at 5 mg/kg,10 mg/ky,30 mg/kg on days 0,1,2 res pectively,and the number and size of astrocyte in or around EAE lesions were ob served with the method of immunohistochemistry.Antibody against glial fibri llary acidic protein(GFAP) was used to demonstrate the astrocyte. Results: Levamisole could increase the incidence of EAE,exacerbate its clinical manifesta tion.On day 16 after immunization,astrocytes in and around the inflamma tory dedmyli native lesion of CNS in EAE rats treated with levamisole increase in number a nd size,of all the effects on incidence,clinical status,and astroglial reactions .Levamisole administered on days 0,1,2 was stronger in effect than administere d on days 0.Modulating the dose of levamisole administered on days 0,1,2,we fou n d the effects of levamisole administered at 10 mg/kg were stronger than administ ered at 5 mg/kg and 30 mg/kg. Conclusion: Levamisole can exacerbate astroglial reaction of central nervous system in EAE .
