摘要
目的 :研究常用抗生素和草酊磷 (Phosphinothricin ,PPT)对盐藻生长的影响。方法 :在固体培养基中加入不同浓度的卡那霉素、青霉素、链霉素、壮观霉素、利福平和PPT ,在液体培养基中加入不同浓度PPT ,对盐藻进行选择培养 ,观察盐藻在选择试剂作用下的生长情况。结果 :卡那霉素、青霉素、链霉素、壮观霉素 16 0 0mg/L时尚不影响固体培养基中盐藻的生长 ;利福平 4 0 0mg/L时可完全抑制固体培养基中盐藻的生长 ;而PPT 3mg/L时即可完全抑制固体和液体培养基中盐藻的生长 ,5mg/L时可以杀死液体培养基中的盐藻。 结论 :PPT对盐藻的生长具有很强的抑制作用 ,在基因工程中 ,以抗除草剂 (BAR)基因作为盐藻的选择标记基因研究盐藻的转化是合适的 。
Aim: To investigate the sensitivity of Dunaliella Salina (D.salina) to the common select reagents. Methods: In the solid and liquid culture media, reagents were added to observe the effect of them on D.salina growth. Results: The study revealed that Kanamycin, Penicillin, Streptomycin or Spectinomycin at 1 600 mg/L could not completely inhibit the growth of D.Salina.However, Rifampin at 400 mg/L could inhibit the grawth of D.salina was most sensitive to Phosphinothricin (PPT), which killed D.salina when PPT was at a concentration of 5 mg/L. Conclusion: PPT should be used when phosphinotricin herbicide resistance (BAR) gene is a selectable marker in the genetic engineering of the D.salina.
