摘要
目的 :研究 p5 3和MGMT基因突变在急性白血病发病中的意义以及 p5 3基因突变与MGMT基因突变的关系。方法 :应用聚合酶链反应 单链构象多态性 (PCR SSCP )技术 ,对 36例急性非淋巴细胞白血病 (AN LL) ,2 6例急性淋巴细胞白血病 (ALL)和 2 3例健康对照组骨髓细胞的 p5 3和MGMT基因突变进行了分析。 结果 :p5 3基因突变率在ANLL中为 16 .7%、ALL中为 19.2 % ,MGMT基因突变率在ANLL中为 13.9%、ALL中为15 .4 % ;6 2例急性白血病中有 4例为 p5 3和MGMT 2个基因同时发生突变 ;正常对照组均未发生p5 3和MGMT基因突变。结论 :p5 3和MGMT基因突变在急性白血病发生与发展中起到一定作用 ,急性白血病中 p5
Aim: To understand the correlation between p53, MGMT gene mutations and acute leukemia, and the correlation between p53 gene mutations and MGMT gene inactivation. Methods: The bone marrow cells of 36 patients with acute non lymphocytic leukemia (ANLL), 26 patients with acute lymphocytic leukemia (ALL) and 23 cases of normal control were detected for mutations in p53 and MGMT gene using polymerase chain reaction single strand conformation polymorphism ( PCR SSCP ). Results: 16.7% of ANLL and 19.2% of ALL contained p53 gene mutations, 13.9% of ANLL and 15.4% of ALL contained MGMT gene mutations, 4 patients in 62 cases of ALL contained both p53 and MGMT gene mutations simultaneously, and neither p53 nor MGMT gene mutation was found in normal control. Conclusion: p53 and MGMT gene mutations may play an important role in the pathogenesis of acute leukemia, and p53 gene mutations were closely correlated with MGMT gene mutations.
