姜黄素拮抗亚砷酸钠所致的DNA链断裂损伤 被引量:11

摘要 目的 观察姜黄素对亚砷酸钠诱发小鼠体内免疫器官细胞DNA链断裂损伤的拮抗作用。方法 用单细胞凝胶电泳法。结果 亚砷酸钠单独染毒后的DNA链断裂损伤的高峰期各器官不同 ,脾脏为染毒后 1h ,胸腺和骨髓在染毒后 3h。姜黄素拮抗亚砷酸钠引起的各器官DNA链断裂损伤的作用在高峰期明显 ,表现为彗星细胞百分比降低 ,并有量效关系。结论 姜黄素可以预防亚砷酸钠诱发的小鼠脾脏、胸腺和骨髓细胞的DNA链断裂损伤。 AIM To investigate the antagonistic effect of curcumin on DNA damage in immune organsin vivo induced by sodium arsenite (NaAsO_2). METHODS Male ICR mice were given curcumin 2, 20 and 200 mg·kg~ -1 for 14 days before treatment with sc injection of NaAsO_2 at the dose of 10 mg·kg~ -1. After injection, the mice were sacrificed at the end of 1 and 3 hours. DNA strand breaks were detected in the splenocyte, thymocyte and marrow cells through single cell gel electrophoresis (SCGE). RESULTS Curcumin was shown to inhibit DNA strand breaks induced by NaAsO_2 in a dose-dependent manner. It was found that curcumin 20 and 200 mg·kg~ -1 could reduce the DNA strand breaks induced by NaAsO_2 1 hour after treatment in cells from spleen. The percent of “comet” cell was decreased. Curcumin 20 and 200 mg·kg~ -1 treatment was shown to reduce the DNA strand breaks induced by NaAsO_2 three hours after treatment in cells from thymus and bone marrow. CONCLUSION Curcumin can prevent the DNA strand breaks induced by NaAsO_2 in cells from spleen, thymus and bone marrowin vivo.
