摘要
目的 探讨热休克反应对由TNF α诱导卵巢癌细胞A2 780凋亡的影响 ,为卵巢癌生物治疗提供实验依据。方法 将体外培养卵巢癌细胞A2 780分为热休克组 (HS)及非热休克组 (NHS)。采用脱氧核苷酸末端转移酶介导的dUTP缺口末端标记法 (TUNEL)和MIT检测肿瘤细胞凋亡指数 (AI)和肿瘤细胞生长抑制率。结果 热休克组细胞凋亡指数比非热休克组细胞凋亡指数明显下降 ,细胞生长抑制率升高 ,差异有显著性意义 (P <0 .0 1) ,且呈剂量依赖效应。结论 热休克反应对由TNF α诱导卵巢癌细胞A2 780凋亡具有抑制作用 。
Objective To explore the resistance effect of HSR for ovarian cancer cells apoptosis induced by TNF α, and provide basic material for biology therapy of ovarian cancer cells. Methods Ovarian cancer cells are divided two groups, heat shock treated and heat shock untreated. TUNEL and MTT are used for examined cell growth inhibition rate and apoptosis index.Results The apoptosis index of HS group are more significantly decreased than the one of untreated HS group ( P <0.01), but the rate of cell growth inhibition of HS are more significantly increased than NHS, which are depended on the dose of TNF α.Conclusion This paper results indicated that HSR has significant inhibition of apoptosis induced by TNF α in ovarian cancer cells, which are depended on the dose of TNF α.
