33份烤烟品种亲缘关系的SSR分析 被引量:4

Genetic Relationship Analysis of 33 Flue-Cured Tobacco Varieties Based on SSR Markers

摘要 本研究利用分布于烟草全基因组24条连锁群上的48对SSR引物,并对33份烤烟品种的亲缘关系进行分析。结果显示:在48个SSR基因座位上共检测获得302个等位基因,每个SSR标记可检测的等位基因数为2~13个,平均6.29个;引物的多态信息含量(PIC)为0.401~0.738,平均0.549,鉴别能力较好;33份烤烟品种间遗传相似性系数(GS)变化范围为0.84~0.96,平均为0.90,表明33份烤烟品种间的遗传差异较小,亲缘关系相对较近。SSR聚类分析表明:在GS分别为0.894和0.910时可清晰地将美国引进及合作选育的27份品种与国内主栽品种区分开,其中,NC-YATAS-7与红花大金元之间及NCT8、NC-YATAS-8与云烟85、云烟87之间的遗传差异性较小,其他24份品种与国内主栽品种间存在一定的遗传差异性。10份合作选育品种和14份引进品种对丰富国内烤烟新选育品种和种质资源的遗传多样性具有重要意义。 In order to investigate the genetic relationship of tobacco germplasm, 33 flue-cured tobacco varieties were analyzed by 48 SSR primer pairs evenly distributed on 24 linkage groups in this study. A total of 302 polymorphic alleles were amplified in 33 flue-cured tobacco varieties, and the number of alleles detected by each SSR primer ranged from 2 to 13, with an average of 6.29 alleles per markers. The PIC(polymorphism information content) values of primers ranged from 0.401 to 0.738, with good identification ability. Genetic similarity coefficient(GS) among the 33 varieties ranged from 0.84 to 0.96, with an average of 0.90, which indicated the genetic variation of the 33 flue-cured tobacco varieties was quite small and the genetic relationship was much near. The result of cluster analysis based on SSR markers showed that 27 US introduced and co-breeding tobacco varieties and 6domestic cultivars could be separated clearly at a relative high level of GS=0.894 and 0.910 respectively, of which,there are small genetic differences between NC-YATAS-7 and Honghuadajinyuan as well as the NCT8,NC-YATAS-8, Yunyan85 and Yunyan87. 10 co-breeding and 14 US introduced tobacco varieties plays an important role in tobacco breeding as well as enriching the genetic diversity of domestic flue-cured tobacco germplasm resources.
