摘要
14-3-3蛋白是真核生物中一类高度保守的调控蛋白,其在植物生长发育、代谢调控、信号转导以及逆境胁迫应答中发挥着非常重要的作用。本研究从棉花纤维均一化cDNA文库中分离得到2个14-3-3基因,分别命名为Gh14-3-3i和Gh14-3-3j。Gh14-3-3i开放阅读框全长789 bp,编码262个氨基酸,Gh14-3-3j开放阅读框全长786 bp,编码261个氨基酸,这2个基因均具有高度保守的14-3-3家族蛋白基序,荧光定量PCR表明,Gh14-3-3i和Gh14-3-3j在棉花纤维、根、茎和叶片中均有表达,并且受到高盐、PEG、ABA等非生物胁迫的诱导表达,本研究可为进一步研究14-3-3蛋白在棉花中的功能提供理论依据。
14-3-3 proteins are highly conserved regulatory proteins in all eukaryotes, plant 14-3-3 s are reported to play important role in development, metabolism, signal transduction and stress response. In this study, two14-3-3 genes were isolated from a cotton fiber normolized cDNA library, which were named Gh14-3-3 i and Gh14-3-3 j. Gh14-3-3 i contained an open reading frame(ORF) of 789 bp, encoded a protein with 262 amino acids, Gh14-3-3 j contained an open reading frame(ORF) of 786 bp, encoded a protein with 261 amino acids, both Gh14-3-3 i and Gh14-3-3 j possess highly conserved 14-3-3 protein motifs. Quantitative RT-PCR analysis showed that Gh14-3-3 i and Gh14-3-3 j expressed in root, stem, leaf and fiber, and were induced by salt, PEG, ABA. Our data will provide theoretical basis for further research on the molecular function of 14-3-3 in cotton.
