摘要
目的建立高效液相色谱法测定牛黄上清丸中胆红素含量的方法。方法以十八烷基键合硅胶为填充剂(Wondasil,4.6 mm×250 mm,5μm),以乙腈-1%冰醋酸溶液(95:5)为流动相,流速为1.0 ml/min,检测波长450 nm,柱温25℃,进样量为10μl。结果胆红素在0.1560~1.560μg/ml范围内线性关系良好(r=0.9999,n=6),平均回收率为100.00%,RSD=1.27%(n=9)。结论该方法准确、简便,可用于牛黄上清丸中胆红素的含量测定。
Objective To establish an HPLC method for the determination of bilirubin in NiuHuang Shangqing Pills.Methods The chromatographic column was loaded with octadecyl silane bonded silica(Wondasil,4.6 mm×250 mm,5μm)with acetonitrile-1%glacial acetic acid(95:5)as mobile phase at the flow rate of 1.0 ml/min.The detection wavelength was 450 nm,the column temperature was 25℃and the injection volume was 10μl.Results The linear range of bilirubin was 0.1560-1.560μg/ml(r=0.9999,n=6).The average recovery was 100.00%with RSD of 1.27%(n=9).Conclusion The method is accurate and simple,and can be adopted in the determination of bilirubin in Niuhuang Shangqing Pills.
