摘要
目的:研究银杏内酯K(GK)能否通过抑制小胶质细胞介导的炎性反应保护髓鞘。方法:C57BL/6雄性小鼠随机分为正常(Normal)组、双环己酮草酰双腙(Cuprizone,CPZ)组和银杏叶内酯K组(GK组),通过免疫荧光染色评估各组髓鞘脱失程度和小胶质细胞的活化情况,Western blot和ELISA检测各组脑组织细胞因子诱导型一氧化氮合酶(iNOS)、精氨酸酶-1(Arg-1)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-10(IL-10)和转化生长因子-β(TGF-β)的表达情况。结果:与正常组相比,CPZ成功诱导了小鼠胼胝体广泛的髓鞘脱失(P<0.001),并引起了小胶质细胞的活化和炎性细胞因子iNOS、TNF-α及IL-6的升高(P<0.001)。与CPZ组相比,GK组有效阻止了髓鞘脱失(P<0.01),抑制了小胶质细胞的活化(P<0.01)。同时GK组下调了促炎细胞因子iNOS、TNF-α和IL-6(P<0.05),上调了抗炎细胞因子Arg-1、IL-10、和TGF-β(P<0.05)。结论:GK通过抑制小胶质细胞介导的炎性反应保护髓鞘。
Objective:The aim of this study was designed to investigate the effect of ginkgolide K(GK)on protection of myelin sheath by inhibiting microglia-mediated inflammatory responses.Methods:C57BL/6 male mice were randomly divided into the normal group,cuprizone(CPZ)group and GK group.Demyelination and activation of microglia were observed by immunofluorescence staining.Cytokines in the brain tissue were detected by Western blot and ELISA,which includes iNOS,Arg-1,TNF-α,IL-1β,IL-6,IL-10 and TGF-β.Results:Compared with the normal group,CPZ successfully induced extensive myelin loss in the corpus callosum of mice in CPZ group(P<0.001),accompanied by the activation of microglia and the increase of iNOS,TNF-αand IL-6(P<0.001,respectively).Compared with the CPZ group,GK effectively pretected myelin loss(P<0.01)and inhibited the activation of microglia(P<0.01).Meanwhile,GK down-regulated the pro-inflammatory cytokines iNOS,TNF-αand IL-6(P<0.05)and up-regulated the anti-inflammatory cytokines Arg-1,IL-10 and TGF-β(P<0.05,respectively).Conclusion:GK may protect myelin sheath by inhibiting microglia-mediated inflammatory responses.
