摘要
目的:观察不同频率疏密波电针“百会”“水沟”对脑缺血再灌注(MCAO/R)恢复期大鼠血脑屏障(BBB)开放效应的影响。方法:将96只成年雄性SD大鼠随机分为假手术组、模型组、2/15Hz组、2/30Hz组、2/50Hz组和2/100Hz组,每组16只。除假手术组外,各电针组和模型组采用改良线栓法建立大鼠MCAO/R模型,各组再随机分成两个亚组,术后饲养3周后实验,各电针组采用相应疏密波频率,电流2mA,持续8min,治疗1次,治疗结束后立即取脑组织,分别采用干湿重法测定大鼠脑组织含水量和荧光分光光度法测定脑组织伊文氏蓝(EB)含量;另将32只大鼠随机分成正常组、模型组、2/50Hz电针组和2/100Hz电针组,每组8只。除正常组外,各组均建立MCAO/R模型,术后饲养3周后实验,各电针组采用相应疏密波频率,电流2mA,持续8min,治疗1次,治疗结束后立即取脑组织,用ELISA检测各组大鼠脑组织神经生长因子(NGF)含量。结果:各组大鼠EB透过量检测显示,除2/30Hz组之外,各电针组EB透过量明显高于模型组与假手术组(P<0.01);2/50Hz、2/100Hz组大鼠的EB透过量高于2/15Hz、2/30Hz组(P<0.05,P<0.01)。各组大鼠脑含水率比较差异无统计学意义。各组大鼠NGF含量检测显示,2/50Hz、2/100Hz组大鼠脑组织NGF含量明显高于正常组(P<0.01),2/100H组大鼠脑组织NGF含量明显高于模型组及2/50Hz组(P<0.01)。结论:百会、水沟穴疏密波电针可促EB透过大鼠血脑屏障,同时也可诱导NGF透过血脑屏障。该效应以2/100Hz疏密波电针最优,且开放效应不会引起大鼠脑水肿。
Objective: To observe the effects of BBB opening in MCAO/Rrecovery rats with electronic acupuncture treatment of different frequency dense wave at Baihui(GV20) and Shuigou(GV26). Methods: A total of 96 adult male SD rats were randomly divided into the sham-operated group, model group, 2/15 Hz electro-acupuncture group, 2/30 Hz electroacupuncture group, 2/50 Hz electro-acupuncture group and 2/100 Hz electro-acupuncture group, 16 rats in each group. Except the sham-operated group, the MCAO/R model of rats was established by modified thread embolism method in each electroacupuncture group and model group. Each group was further divided into two subgroups randomly. After 3 weeks of feeding,the experiment was conducted. The corresponding dense wave frequency was used in each electro-acupuncture group, current of 2 mA, lasted for 8 minutes, and treated once. Brain tissue was taken immediately after treatment. Brain water content and EB content were determined by dry-wet weight method and fluorescence spectrophotometry, respectively. A total of 32 rats were randomly divided into control group, model group, 2/50 Hz electro-acupuncture group and 2/100 Hz electro-acupuncture group,with 8 rats in each group. MCAO/R model was established in all groups except the control group. After 3 weeks of feeding, the experiment was conducted. The corresponding dense wave frequency is used in each electronic acupuncture group, current 2 mA,lasted for 8 minutes, and treated once. Brain tissue was taken immediately after treatment. The content of NGF in brain tissue of each group was detected by ELISA. Results: Except the 2/30 Hz electro-acupuncture group, the penetrationamounts of EB in each electro-acupuncture group was significantly higher than that in model control group and sham-operated group(P<0.01). The penetrationamounts of EB in 2/50 Hz and 2/100 Hz electro-acupuncture group was higher than that in 2/15 Hz and 2/30 Hz electroacupuncture group(P<0.05, P<0.01). There was no significant difference in brain water content among six groups. The content of NGF in brain tissue of rats in 2/50 Hz and 2/100 Hz EA groups was significantly higher than that in normal group(P<0.01), the content of NGF in brain tissue of rats in 2/100 Hz EA group was significantly higher than that in the model group and 2/50 Hz EA group(P<0.01). Conclusion: Dilatational waves of electronic acupuncture can have the effect of promoting EB through the BBB at GV20 and GV26, and NGF can also be induced through the BBB. This effect is optimized at 2/100 Hz dilatation wave of electronic acupuncture, and the opening effect don’t cause cerebral edema in rats.
