摘要
研究表明慢性炎症与肿瘤的发生发展密切相关。慢性炎症主要通过内源性途径和外源性途径参与肿瘤的进展,转录因子NF-кB和STAT3等在其中起着重要的作用。胰腺导管腺癌(PDAC)是恶性程度最高的肿瘤之一,慢性炎症可协同原癌基因的突变促进PDAC发生发展。在慢性炎症恶性转化为PDAC的过程中,炎症相关信号传导通路间的crosstalk发挥了调控作用,其中NF-кB和STAT3是两个重要关键调控点。基于信号通路间的crosstalk以关键调控点为靶点,或可为PDAC的预防和治疗提供新的思路。本文就慢性炎症在肿瘤及PDAC发生发展中的作用、慢性炎症引发PDAC相关炎症信号网络以及NF-кB和JAK/STAT3两条信号通路间的相互作用等研究动态进行简要综述。
Studies have demonstrated a strong correlation between chronic inflammation and tumor initiation and progression.Chronic inflammation is mainly involved in tumor progression through intrinsic and extrinsic pathways,and transcription factors such as NF-κB and STAT3 play important roles.Pancreatic ductal adenocarcinoma(PDAC)is one of the most lethal malignancies.Chronic inflammation can promote the development of PDAC in cooperation with mutation of protooncogene.Inflammation signaling pathway plays an important role in malignant transformation in the PDAC,in which NF-κB and STAT3 are two major transcription factors.Targeting the key regulator points based on the crosstalk between inflammation signaling and PDAC may potentially represent a novel for PDAC prevention and treatment.This article briefly reviews the role of chronic inflammation in the development of PDAC,the PDAC-related inflammatory signal network caused by chronic inflammation,and crosstalk between NF-кB and JAK/STAT3 signaling pathways.
