镉胁迫对食蚊鱼肝组织基因表达的影响 被引量:3

摘要 以食蚊鱼为受试生物,采用半静水式亚急性毒性试验法以及荧光定量PCR技术,分别测定镉胁迫后食蚊鱼肝组织中热休克蛋白70(HSP70)、热休克蛋白90(HSP90)、细胞色素氧化酶1A(CYP1A)以及金属硫蛋白(MT)基因表达水平的影响,以探讨镉胁迫对食蚊鱼的毒性效应,了解重金属镉在水体中的毒性,拓展食蚊鱼在环境毒理科学研究中的应用范围。结果表明,随着暴露时间的延长,肝组织中HSP70基因和CYP1A基因相对表达水平表现为逐渐下调的趋势,HSP90基因和MT基因表达水平先显著上调后逐渐降低,且趋于稳定。本研究通过多个角度综合探讨镉对食蚊鱼的毒性作用机制,将为防治镉污染提供科学依据,拓展食蚊鱼在环境毒理科学研究中的应用范围,也为其成为模式生物提供数据支持。 The gene expression patterns of heat shock protein 70(HSP70),heat shock protein 90(HSP90),cytochrome oxidation 1A(CYP1A),and metallothionein(MT)were investigated in liver of mature mosquitofish Gambusia affinis with body weight of(2.80±0.05)g exposed to Cd^2+ concentrations of 0,0.564,1.128 and 2.255 mg/L(0,1/40,1/20 and 1/10 of 96 h median lethal concentration)using semi-hydrostatic toxicity test method for 1,3,5,and 7 d at water temperature of(24±2)℃by fluorescent real-time quantitative PCR to study the effect of cadmium(Cd^2+)in water environment on the gene mRNA expression levels.The results showed that the expression patterns of HSP70 and MT in the liver were induced at first and then inhibited with the increment in exposure period,with significantly up-regulated HSP90 with the increment in exposure concentrations at the same exposure time.The CYP1A expression levels were significantly inhibited with the increment in exposure period,which implies the potential negative effect of Cd^2+ on physiological status of mosquitofish as evidenced by changing of the genes expression patterns.The findings comprehensively explore the toxic mechanism of cadmium in mosquitofish and provide a scientific basis for the prevention and control of cadmium pollution.
