不同纯化方法对补阳还五汤水提液物质组成的影响 被引量:3

摘要 目的基于溶液环境-化学成分分析比较不同分离纯化方法对补阳还五汤水提液物质组成的影响。方法以补阳还五汤水提液为实验体系,分别测定高速离心法、微滤法、醇沉法及絮凝法等不同纯化方法处理前后原液和纯化液的溶液环境中pH值、电导率、浊度及黏度等物化参数;分析蛋白质、鞣质、淀粉及果胶等高分子物质含量变化;测定多糖、黄芪甲苷及毛蕊异黄酮葡萄糖苷等成分的含量。结果补阳还五汤不同纯化方法的溶液环境中,物理参数浊度基本吻合各高分子含量的变化规律,高分子去除率显示最优纯化方法为微滤法,其次为醇沉法;而各指标成分提示醇沉法保留率最高,微滤法最差。结论醇沉法对补阳还五汤的纯化具有适宜性。 Objective Based on the analysis of solution environment-chemical composition,the effects of different separation and purification methods on the composition of extracts from Buyanghuanwu Decoction were compared.Methods Taking Buyanghuanwu Decoction as the test system,the physicochemical parameters such as pH,conductivity,turbidity and viscosity in the original solution and the purified solution before and after treatment by different purification methods such as high-speed centrifugation method,microfiltration method,alcohol precipitation method and flocculation method were measured respectively.The content of protein,tanning,starch and pectin etc.were analyzed.The contents of polysaccharides,astragalus methyl glycoside and triticum isoflavone glucoside were determined.Results In the solution environment of different purification methods of Buyanghuanwu Decoction,the physical parameter turbidity basically accords with the change rule of each polymer content.The polymer removal rate shows that the optimal purification method is microfiltration method,followed by alcohol precipitation method;it is suggested that the alcohol precipitation method has the highest retention rate and the microfiltration method is the lowest.Conclusion Alcohol precipitation method is suitable for the purification of Buyanghuanwu Decoction.
