摘要
目的建立用于金莲花药材质量评价的含量测定方法,对药材质量进行初步评价,优化金莲花的提取工艺。方法采用HPLC法和紫外-可见分光光度法分别测定金莲花中荭草苷、牡荆苷和总黄酮的含量。色谱柱为Shim-pack VP-ODS C 18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-4 mL·L^-1磷酸溶液(14∶86);流速为1.0 mL·min^-1;柱温为30℃;检测波长为340 nm。采用紫外-可见分光光度法测定总黄酮含量。采用L 9(3 4)正交实验法优化金莲花的提取工艺,并进行验证实验。结果荭草苷、牡荆苷和总黄酮分别在15.92~95.55,16.98~101.91和8.128~48.768μg·mL^-1范围内线性关系良好(r值分别为0.999 8,0.999 7和0.999 7);平均加样回收率分别为100.10%,100.81%和100.17%;RSD值分别为1.69%,1.46%和1.60%(n=6);精密度、稳定性和重复性实验的RSD值均小于2.0%,样品溶液在12 h内稳定;最优提取工艺条件:加10倍量体积分数为70%的乙醇,提取2次,每次1.0 h。结论该方法简便、准确、重复性好,可用于金莲花药材的质量评价和控制;优化后的提取工艺稳定、可行。
Objective To establish a method for the quality evaluation of the medicinal materials of Trollius chinensis,to evaluate the quality of the medicinal materials and to optimize the extraction process.Methods HPLC and UV-Vis spectrophotometry were used to determine the content of orientin,vitexin and total flavonoids.The column was Shim-pack VP-ODS C 18(250 mm×4.6 mm,5μm).The mobile phase was acetonitrile-4 mL·L^-1 phosphoric acid solution(14∶86),the flow rate was 1.0 mL·min^-1,the detection wavelength was set at 340 nm and the column temperature was 30℃.The total flavonoids were determined by UV-vis spectrophotometry.An L 9(3 4)orthogonal experiment was used to optimize the extraction process of Trollius chinensis and a validation test was carried out.Results The method had good linear relationship within the ranges of orientin,vitexin and total flavonoids were 15.92-95.55(r 1=0.999 8),16.98-101.91(r 2=0.999 7)and 8.128-48.768μg·mL^-1(r 3=0.999 7),the average recoveries were 100.10%(RSD=1.69%),100.81%(RSD=1.46%)and 100.17%(RSD=1.60%)(n=6).RSDs of precision,stability and reproducibility tests were lower than 2.0%.The sample solution was stable within 12 h.The optimized ethanol extraction conditions were as follows:extracted 2 times with 10 times of the amount of 70%ethanol,1.0 h.Conclusion The method is simple,accurate and repeatable,and can be used for the quality evaluation and control of medicinal materials.The optimized extraction process is stable and feasible.
