摘要
采用白瓷板比色法对砷耐受菌Pseudomonas taiwanensis 11(P11)的氧化As(Ⅲ)或还原As(Ⅴ)能力进行鉴定。结果表明,P11对As(Ⅲ)具有显著的氧化能力,As(Ⅲ)被氧化量的增加与培养时间呈正相关,对As(Ⅴ)没有明显的还原能力。P11培养6.5 h,可将30%的As(Ⅲ)氧化为As(Ⅴ);P11培养9.0 h,可将70%的As(Ⅲ)氧化为As(Ⅴ)。与高效液相色谱法(HPLC)和电感耦合等离子体原子发射光谱法(ICP-AES)相比,该方法操作简单、反应迅速、成本低,是一种快速、直观高效的鉴定砷耐受菌的方法。
We identified the oxidation ability of arsenic-tolerant bacterium Pseudomonas taiwanensis 11(P11)for As(Ⅲ),and the reduction ability for As(Ⅴ).The results show that P11 has significant oxidation ability to As(Ⅲ),and the increase of As(Ⅲ)oxidation content is positively correlated with culture time.While P11 has no obvious reduction ability to As(Ⅴ).Moreover,after being cultured for 6.5 h,P11 can oxidize approximate 30%of As(Ⅲ)into As(Ⅴ),and after being cultured for 9.0 h,P11 can oxidize approximate 70%of As(Ⅲ)into As(Ⅴ).Compared with high performance liquid chromatography(HPLC)and inductively coupled plasma-atomic emission spectrometry(ICP-AES),white porcelain plate colorimetry has the characteristics of simple operation,quick response,and low cost,which is a rapid,visual,and efficient method for the identification of arsenic-tolerant bacteria.
