摘要
目的检测大鼠同种异体角膜移植术后,随着排斥反应发生,不同时间外周血中自然杀伤(nature killed,NK)细胞计数的变化,初步揭示NK细胞在角膜移植排斥反应早期的作用。方法选用远交系Wistar大鼠角膜为受体,以SD大鼠角膜为供体建立同种异体角膜移植模型为实验组(20只);自体角膜移植为对照组(12只)。于术后第2、4、6、8、10、12天,断尾取其外周血,进行NK细胞的流式细胞计数,观察术后不同时间外周血中NK细胞占淋巴细胞比例的变化。结果在实验组术后早期大鼠外周血中NK细胞占淋巴细胞比例明显增高于对照组(P<0.05),NK细胞计数峰值出现在术后第(4.40±0.82)d。结论NK细胞在大鼠同种异体角膜移植排斥反应发生过程中,通过其直接杀伤以及抗原提呈等方式,在早期的角膜移植排斥反应中起到一定作用,为后续排斥反应启动机制的进一步研究提供了基础。
Objective To detect changes in natural killer(NK)cell count in the peripheral blood,with the development of rejection after rat corneal allograft,and initially reveal the role of NK cells in rat corneal allograft rejection.Methods An appropriate animal model of corneal allograft rejection with Wistar rats as the receptor and SD rats as the donator was established.At the time points of PO2,PO4,PO6,PO8,PO10 and PO12,20 and 12 rats were respectively designated as the experimental group and the control group.Peripheral blood was taken from the cut rat tails from the control group,and NK cell count was calculated by flow cytometry,and then,detections were made on the changes of NK cell count in the ratio of lymphocytes at different time points after surgery.Results The proportion of NK cells in lymphocytes of the corneal allograft group was significantly higher in the initial stage after surgery than that of the control group(P<0.05).The peak of NK cell counts appeared(4.40±0.82)days after surgery.Conclusion NK cells displayed a certain role in the process of early corneal allograft rejection through direct killing and antigen presentation.The study results might provide a basis for further research on the trigger mechanism in the early corneal allograft rejection.
