垂体瘤患者外周血T细胞和NK细胞LAG3表达的研究 被引量:2

摘要 目的:检测垂体瘤患者外周血T细胞和自然杀伤细胞(NK)的比例及其表面LAG3的表达,并探讨其表达改变的意义。方法:收集30例脑垂体瘤患者术前和30例健康志愿者外周血标本,流式细胞术检测CD3^+CD4^+T、CD3^+CD8^+T、CD3^-CD56^+NK细胞比例及细胞表面LAG3的表达。结果:与健康志愿者相比,垂体瘤患者外周血CD4+T细胞比例增加、NK细胞比例降低,差异有统计学意义(P=0.005 7;P<0.001),而CD8^+T比例无显著差异(P=0.442 5);垂体瘤患者CD4^+T、CD8^+T和NK细胞表面LAG3分子的表达水平均显著升高(P=0.001 8、P<0.001、P<0.001)。结论:垂体瘤患者外周血T细胞和NK细胞表面LAG3表达水平均升高,提示垂体瘤可能通过诱导LAG3表达上调,参与介导T细胞和NK细胞免疫耐受,促进肿瘤进展。 Objective:To analyze the expression of negative costimulatory molecule lymphocyte activation gene 3(LAG3)on peripheral blood T cells and natural killer(NK)cells from patients with diagnosis of pituitary adenomas and its significance.Methods:Peripheral blood samples were collected from 30 pituitary adenomas patients and 30 healthy subjects(HC).The frequencies of CD3^+CD4^+T cells、CD3+CD8+T cells and CD3^-CD56^+NK cells were examined by flow cytometry.Simultaneously,the expression of LAG3 was detected.Results:The proportion of CD3^+CD4^+T cells in pituitary adenomas was significantly higher than that in the healthy control(P=0.005 7),while the proportion of CD3^-CD56^+NK cells was significantly decreased(P<0.001).Additionally,there was no statistical difference on CD3^+CD8^+T cells(P=0.442 5).The expression of LAG3 on CD3^+CD4^+T cells,CD3^+CD8^+T cells and CD3^-CD56^+NK cells in pituitary adenomas was significantly higher than that in healthy control(P=0.001 8;P<0.001;P<0.001).Conclusion:The expression of LAG3 on peripheral blood T cells and NK cells was significantly increased,which may mediate peripheral immune tolerance and discount any effects of anti-tumor immunity,participating in the generation and progression of pituitary adenomas.
