摘要
牛星状病毒(BoAstV)ORF2基因在感染与免疫过程中发挥重要作用,为了解BoAstV在川西北牦牛中的流行及ORF2基因的分子特征,采集2016年-2017年川西北135份牦牛腹泻粪便样本,采用RT-PCR方法对临床样本进行检测,并对阳性样本进行完整ORF2基因的克隆。结果显示,从135份样本中检出7份BoAstV阳性,检出率为5.2%;获得的2条完整ORF2基因长度皆为2 313 bp,与GenBank中仅有的1条牦牛源BoAstV S8株的ORF2完整基因的氨基酸同源性分别为89.4%和86.1%,与BoAstV ORF2氨基酸同源性为28.5%~52.2%;这2条序列在进化树上与牦牛源BoAstV S8株单独聚于一支,与BoAstV有明显的遗传距离。研究结果丰富了牦牛源BoAstV ORF2基因信息,为研究牦牛源BoAstV的诊断和防控提供参考。
Bovine astrovirus(BoAstV)ORF2 plays an important role in infection and immunity.The purpose of this experiment was to investigate the prevalence of BoAstV from yaks and to explore the molecular characteristics of ORF2 in northwest Sichuan.A total of 135 yak diarrhea samples were collected in northwestern Sichuan from 2016 to 2017.The clinical samples were detected by the specific RT-PCR method for detecting BoAstV in yaks,and the complete ORF2 were cloned from AstV positive samples.The results showed that 7 samples were detected BoAstV as positive from 135 samples,and the detection rate was 5.2%.Two complete ORF2 gene sequences were obtained,which both are 2 313 bp in length.The amino acid homologies of the two complete ORF2 gene sequences obtained were 89.4%and 86.1%respectively with the only one complete ORF2 from yak BoAstV S8 strain in GenBank,and 28.5%-52.2%amino acid homology with BoAstV ORF2 in GenBank.These two sequences were clustered together in a phylogenetic tree with yak BoAstV S8 strain,and had obvious genetic distance from BoAstV.In summary,two complete sequences of yak BoAstV ORF2 were obtained,which contributed to genic information of ORF2 of BoAstV in yaks,also provided a reference for diagnosis and prevention of yak BoAstV.
