摘要
为研究青岛百合花发育问题的分子调控机理,以青岛百合花朵为材料,克隆得到一个MADS-box家族E类基因SEPALLATA3(SEP3),命名为LtSEP3,开放阅读框为687 bp,预测编码228个氨基酸。生物信息学分析结果表明,LtSEP3蛋白的相对分子质量为26 179.58 D,理论等电点在8.68,不稳定系数为33.66,属于稳定蛋白,总平均亲水性(GRAVY)为-0.819,脂肪系数为79.96。氨基酸序列比对显示,LtSEP3基因编码的蛋白具有典型的MADS结构域和K-box区。LtSEP3与同为百合属植物的岷江百合的MADS蛋白的氨基酸序列亲缘关系最近;qRT-PCR分析显示,LtSEP3具有组织表达特异性,主要在花中表达,在鳞茎、茎、叶中几乎不表达,在花朵开放的中后期,LtSEP3表达量达到峰值。本研究认为,LtSEP3基因可能在青岛百合花朵开放和花发育过程中具有重要作用。
In order to study the molecular regulation mechanism of flower development in Lilium tsingtauense,a SEPALLATA3(SEP3)gene was cloned from petals by RT-PCR and RACE,named LtSEP3,which is a member of E class of MADS-box family.The open reading frame of LtSEP3 is 687 bp and it predicts to encode 228 amino acids.Bioinformatics analysis showed that the relative molecular weight of LtSEP3 is 26 179.58 D,the theoretical isoelectric point is 8.68 and the instability coefficient is 33.66.LtSEP3 belongs to stable protein.The total average hydrophilicity(GRAVY)is-0.819 and the fat coefficient is 79.96.Amino acid sequence alignment showed that the protein encoded by the LtSEP3 had a typical MADS domain and K-box region;LtSEP3 is closely related to the amino acid sequence of MADS protein of Lilium regale.LtSEP3 was expressed mainly in flowers while it could not be detected in bulbs,stems and leaves.LtSEP3 expression reached its peak when the flower was fully expanded.This study suggests that LtSEP3 gene may play an important role in flower opening and flower development of Lilium tsingtauense.
