摘要
目的:探讨糖尿病大鼠血清睾酮(T)、性激素结合蛋白(SHBG)水平与阴茎勃起功能障碍(ED)的关系及其发生的可能机制。方法:将43只雄性Sprague-Dawley大鼠随机分为对照组(n=14)和糖尿病组(n=29),糖尿病组喂食高脂肪饮食24周后,腹膜腔内注射小剂量的链脲佐菌素(STZ)20 mg/kg,构建2型糖尿病模型,对照组给予普通饲料及注射相同剂量的柠檬酸钠缓冲液。造模两周后,两组均皮下注射适量阿扑吗啡(APO)溶液,诱导阴茎勃起,记录大鼠阴茎勃起频率和勃起率;测定大鼠血清T和SHBG水平;苏木精-伊红(HE)染色及EVG染色观察两组大鼠阴茎组织的病理变化。结果:与对照组相比,糖尿病组勃起频率、勃起率及血清SHBG和T水平均显著降低(P<0.05)。阴茎HE染色显示:与对照组相比,糖尿病组大鼠阴茎海绵体平滑肌分布不均匀,排列紊乱、松散,阴茎海绵体内海绵窦数量明显减少;阴茎EVG染色显示:与对照组相比,糖尿病组大鼠阴茎海绵体弹性纤维向外突出,平滑肌纤维分布面积减小,胶原纤维明显增多。结论:糖尿病可影响大鼠体内T和SHBG的分泌和代谢,促使阴茎组织发生病理改变,导致ED。
Objective:To investigate the relationship between the serum levels of testosterone(T)and sex hormone binding protein(SHBG)and erectile dysfunction(ED)in diabetes rats,and explore its possible mechanism.Methods:A total of 43 male SD rats were randomly divided into a control group(n=14)and a diabetic group(n=29).The diabetic group rats were fed with high lipid diet for 24 weeks and then injected with low-dose of streptozotocin(STZ,20 mg/kg),in order to established the rat models of type 2 diabetes.Meanwhile,the rats in the control group were fed with normal diet and intraperitoneally injected with the equal volume of sodium citrate buffer.Two weeks after modeling,the apomorphine(APO)was injected subcutaneously to induce penile erection.The erectile frequency and erection rate were recorded.Serum T and SHBG levels were measured.Hematoxylin-eosin(HE)staining and EVG staining were used to observe the pathological changes of penile tissues.Results:Compared with the control group,the erectile frequency and erection rate in diabetic group were significantly lower than those in control group(P<0.05).HE staining showed uneven distributed sponge smooth muscle,disordered arrangement and loose as well as significantly reduced number of cavernous sinus in penis sponge body in diabetes group.EVG staining indicated that the elastic fibers of the penile cavernous body protruded outward,the distribution area of smooth muscle fibers decreased,and the collagen fibers increased significantly in the diabetic group.Conclusion:Diabetes may affect the T and SHBG secretion and metabolism,promote the pathological changes of penile tissue,leading to ED.
