天然冰片对局灶性脑缺血模型大鼠的脑保护作用研究 被引量:11

Brain Protection of Natural Borneol on Focal Cerebral Ischemia in Model Rats

摘要 目的:考察天然冰片对局灶性脑缺血大鼠的脑保护作用。方法:SD大鼠随机分为假手术组、模型组、吐温(溶剂)+模型组、尼莫地平10. 8 mg/kg组、天然冰片0. 15、0. 3、0. 6 g/kg 7组。预给药三天后,采用线栓法致大脑中动脉栓塞造成局灶性脑缺血模型,记录大鼠翻正反射消失到苏醒的时长;参照Zea Longa评分法对大鼠神经功能损伤进行评分;采用干湿重法测得脑指数、Δ脑指数和脑组织含水率;采用TTC染色法染色观测缺血侧大脑的梗死灶,并计算脑梗死率;采用HE染色法观察大鼠缺血半暗带脑组织形态。结果:与假手术组比较,模型组和溶媒模型组苏醒时间、神经功能评分、脑组织含水率、脑指数、脑梗死率和脑组织病理评分明显增加或增高,大鼠海马区损伤明显。天然冰片0. 15、0. 3、0. 6 g/kg剂量组均能显著延长麻醉后p MCAO模型大鼠的苏醒时间;能明显降低大鼠行为评分;并能明显降低p MCAO模型大鼠Δ脑指数、脑组织含水率以及脑梗死率;天然冰片0. 3、0. 6 g/kg剂量组能有效改善p MCAO模型大鼠海马区脑细胞损伤。结论:天然冰片能够预防局灶性脑缺血发生而发挥脑保护作用,其中0. 6 g/kg剂量组的作用最明显。 Objective: In this search,we investigated the brain protective effect of natural borneol on rats with focal cerebral ischemia. Methods:SD rats were randomly divided into 7 groups: the sham operation group,the model group,twain (solvent) + model group,10. 8 mg/kg nimodipine (positive drug) group and natural borneol groups (0. 15、0. 3、0. 6 g/kg). After three days of pre-administration,focal cerebral ischemia model was induced by embolization of middle cerebral artery by thread occlusion. The duration time from the disappearance of righting reflex to the revival in each group of rats was recorded. Zea Longa scoring method was used to grade neurological injury in model rats. Dry-wet weight method was used to measure cerebral index,Δcerebral index and brain edema rate. The rate of cerebral infarction was calculated by TTC staining method. The morphology of ischemic penumbra was observed by HE staining. Results: Compared with the sham operation group,the awaken time,neurological injury score,cerebral edema rate,cerebral index,cerebral infarction and pathological injury scores were increased in both model group and twain (solvent) + model group,also the damage in hippocampus was increased. 0. 6,0. 3 and 0.15 g/kg natural borneol significantly prolonged the waking time of pM CAO model rats after anesthesia. The behavioral score of rats was decreased. The cerebral water content,cerebral edema rate and cerebral infarction rate in the ischemic side of pM CAO model rats were significantly reduced. The damages of brain cells in the hippocampus were improved by 0. 6 and 0. 3 g/kg natural borneol. Conclusion: Natural borneol has the function of preventing focal cerebral ischemia,especially at the dose of 0. 6 g/kg.
