摘要
目的研究补骨脂抗抑郁有效成分群(effective components group of fructus psoraleae,EGFP)的抗抑郁药效及其安全性。方法补骨脂通过乙醇提取,石油醚萃取后得到含有补骨脂总黄酮、补骨脂定及补骨脂酚的有效成分群,其中不含补骨脂素及异补骨脂素,并控制补骨脂酚的含量。选取该EGFP、阳性对照品氟西汀和文拉法辛给予雌性ICR(Institute of Cancer Research)小鼠。7天后检测行为受限后的不动时间及血清和脑内海马组织的(5-羟色胺,5-hydroxytryptamine)含量;采用急性毒性及重复给药试验研究该有效成分群的安全性。结果EGFP可降低小鼠行为受限的不动时间(P<0.05,P<0.01),增加血清及海马组织中5-HT含量(P<0.05),且具有剂量依赖性;该有效成分群对小鼠的最大给药量为7 g·kg^-1,300 mg·kg^-1给药56 d后,血清生化及组织病理学检查未见明显异常。结论EGFP具有抗抑郁作用,无明显毒性。
Objective:To study the antidepressant efficacy of the effective components group of Fructus Psoraleae(EGFP)foranti-depression anditssafety.Method:The EGFP was obtained after EtOH extraction and petroleum ether extraction,which contained total flavonoid,psoralidin,and bakuchiol.There were nopsoralen andisopsoralen of theEGFP,andthe content of bakuchiol was controlled.The EGFP and positive control of fluoxetine and venlafaxine were employed to treat female ICR mice.Inactive time after limitation of activity and content of 5-HT in serum and hippocampal tissueafter 7 d were detected.The safety of this EGFP wasstudied by acute toxicity and repeatadministration tests.Results:The EGFP canreduce the inactive time after limitation of activity(P<0.05,P<0.01)and increase the content of 5-HT in serum and hippocampal tissue,withitis dose-dependent(P<0.05).The maximum doseof EGFP to mice was 7 g·kg^-1.No obvious abnormality was found in serum biochemical and histopathological examinations after treating with 300 mg·kg^-1 EGFP for 56 d.Conclusion:The EGFP possesses antidepressant efficacy and no obvious toxicity.
