摘要
目的建立蒙药拉哈如各贡斯勒散的定性鉴别及含量测定方法。方法采用TLC法对方中栀子苷进行定性鉴别。运用HPLC法对紫草中左旋紫草素进行含量测定,色谱条件为Kromasil C 18色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.025 mol·L-1磷酸溶液(85∶15)为流动相;检测波长为516 nm;流速为1.0 mL·min^-1;进样量为20μL;柱温为30℃。结果薄层色谱中栀子苷斑点清晰,重复性好,易于鉴别。左旋紫草素质量浓度在0.61~39.00μg·mL-1范围内与峰面积线性关系良好(r=0.9999),平均回收率为98.3%(RSD=2.73%)。结论该法操作简便可行、重复性好、准确度高,可用于拉哈如各贡斯勒散的质量控制。
Objective To establish a method for qualitative and quantitative analysis of Mongolian medicine Laharugegongsile Powder.Methods Geniposide in the preparation was identified by TLC.Shikonin in Radix Shikonin was determined by HPLC.The HPLC analysis was performed on a Kromasil C 18 column(250 mm×4.6 mm,5μm)with methanol-0.025 moL·L-1 phosphoric acid(85∶15)as the mobile phase.The detection wavelength was 516 nm,the flow rate was 1.0 mL·min^-1,the sample injection volume was 20μL,and the column temperature was 30℃.Results TLC identification spots were clear,reproducible and geniposide can be identified easily.HPLC method showed good linear relationship in the range of 0.61-39.00μg·mL-1 for shikonin(r=0.9999),and the average recovery was 98.3%(RSD=2.73%).Conclusion The established method is simple,reliable,reproductive and accurate.The method can be used for the control quality of Laharugegongsile Powder.
