应用正交实验优化扁藻胞外多糖硒化工艺的研究 被引量:3

摘要 目的探讨正交实验优化扁藻胞外多糖的硒化工艺。方法低温蒸馏浓缩扁藻培养液,分离、纯化扁藻胞外多糖,以正交实验分析影响扁藻胞外多糖硒化的因素,分别检测扁藻胞外多糖和硒化扁藻胞外多糖的紫外吸光度和红外吸光度。结果正交实验显示最佳多糖硒化工艺为:反应时间为8 h,硝酸质量分数为3%,多糖和亚硒酸钠的质量比为1:1.2,超声功率为70 W。扁藻胞外多糖与硒化扁藻胞外多糖样本经紫外线检测提示均无蛋白质杂质,红外吸光度扫描显示扁藻胞外多糖为吡喃型多糖,以共价键形式连接的硒化多糖修饰成功。结论以正交实验统计方法筛选扁藻胞外多糖硒化工艺操作简单、方便。 Objective To optimize the selenization process of extracellular polysaccharides of Platymonas using orthogonal experiment.Methods Extracellular polysaccharides were cryodistilled,isolated and purified from the culture medium of Platymonas platensis.The influencing factors for extracellular polysaccharide selenization of Platymonas were analyzed by orthogonal experiment.Ultraviolet and infrared absorbances of extracellular polysaccharides of Platymonas with or without selenide were determined.Results Orthogonal experiments showed that the optimum selenization processes of polysaccharide were 8 h reaction time,3% nitric acid,1:1.2 of polysaccharides/sodium selenite,70 W ultrasound.Ultraviolet detection found no protein impurities in extracellular polysaccharides of Platymonas.Infrared absorbance scanning showed that the extracellular polysaccharides of Platymonas were pyran.The selenide polysaccharide was successfully modified by covalent bond.Conclusion The orthogonal experiment is simple and convenient to optimize the selenization process of extracellular polysaccharides of Platymonas.
