丙泊酚抑制胶质瘤细胞侵袭和转移的内质网应激机制 被引量:1

Endoplasmic reticulum stress mechanism of propofol inhibiting invasion and metastasis of glioma cells

摘要 目的观察丙泊酚对胶质瘤细胞侵袭和转移的影响,探讨内质网应激(ERS)在胶质瘤细胞侵袭和转移的作用机制.方法人胶质瘤细胞系U87-MG细胞培养分为对照组(C组)、2μg/ml组、5μg/ml组和10μg/ml组,分别加入浓度为0、2、5和10μg/ml丙泊酚溶液.采用MTT法检测细胞活力;Transwell侵袭迁移测定侵袭细胞数;划痕实验测定迁移率;Western blot检测细胞葡萄糖调节蛋白78(GRP78)和CCAAT/增强子结合蛋白同源蛋白(CHOP)表达.比较每组细胞细胞活力、侵袭细胞数、迁移率及GRP78和CHOP表达.结果2、5和10μg/ml组胶质瘤细胞活力低于C组,且随丙泊酚剂量增加胶质瘤细胞活力及迁移率逐渐降低,差异有统计学意义(P<0.05).2、5和10μg/ml组胶质瘤细胞侵袭细胞数少于C组,2μg/ml组胶质瘤细胞侵袭细胞数多于5、10μg/ml组,差异有统计学意义(P<0.05).C组与5、10μg/ml组胶质瘤细胞GRP78表达比较,差异有统计学意义(P<0.05).2μg/ml组与5、10μg/ml组胶质瘤细胞GRP78表达比较,差异有统计学意义(P<0.05).5μg/ml组与10μg/ml组胶质瘤细胞GRP78表达比较,差异有统计学意义(P<0.05).C组与2、5和10μg/ml组胶质瘤细胞CHOP表达比较,差异有统计学意义(P<0.05).10μg/ml组与2、5μg/ml组胶质瘤细胞CHOP表达比较,差异有统计学意义(P<0.05).结论丙泊酚可抑制人胶质瘤细胞的侵袭和迁移能力,其机制可能与激活内质网应激标志蛋白GRP78和CHOP表达变化有关. Objective To observe the effect of propofol on invasion and metastasis of glioma cells,and discuss the mechanism of endoplasmic reticulum stress(ERS)in invasion and metastasis of glioma cells.Methods Human glioma cell line U87-MG was divided into control group(group C),2μg/ml group,5μg/ml group and 10μg/ml group,and propofol solution was added at concentrations of 0,2,5 and 10μg/ml,respectively.Cell viability was detected by MTT assay,number of invasive cell by Transwell invasion and migration assay,migration rate by scratch assay,and expression of cell glucose regulatory protein 78(GRP78)and CCAAT/enhancer binding protein homologous protein(CHOP)by Western blot.Cell viability,number of invasive cell,migration rate and expression of GRP78 and CHOP were compared in each group.Results The cell viability of glioma cells in 2,5 and 10μg/ml group was lower than that in group C,and the cell viability and migration rate of glioma cells decreased with the increase of propofol dosage,the difference was statistically significant(P<0.05).The number of invasive cell of glioma cells in 2,5 and 10μg/ml group was lower than that in group C,and 2μg/ml group was more than 5 and 10μg/ml group.Their difference was statistically significant(P<0.05).There was statistically significant difference in expression of GRP78 of glioma cells in group C and 5 and 10μg/ml group(P<0.05).2μg/ml group and 5 and 10μg/ml group had statistically significant difference in expression of GRP78 of glioma cells(P<0.05).5μg/ml group and 10μg/ml group had statistically significant difference in expression of GRP78 of glioma cells(P<0.05).Group C and 2,5 and 10μg/ml group had statistically significant difference in expression of CHOP of glioma cells(P<0.05).10μg/ml group and 2,5μg/ml group had statistically significant difference in expression of GRP78 of glioma cells(P<0.05).Conclusion Propofol inhibits the invasion and migration of human glioma cells,and its mechanism may be related to the activation of endoplasmic reticulum stress marker proteins GRP78 and CHOP.
