摘要
[目的]观察川骨片治疗家兔激素性股骨头缺血性坏死的作用机制.[方法]将56只日本雄性大耳兔,随机分为空白组(16只)和造模组(40只).采用贺西京法复制家兔激素性股骨头缺血性坏死模型,6周后处死空白组4只和造模组4只,进行模型鉴定,并计算空骨陷窝率.造模成功后,将造模组随机分为模型对照组、川骨片治疗组、仙灵骨葆胶囊治疗组,每组各12只.空白组不予特殊处理,其余3组分别对应给予蒸馏水、川骨片、仙灵骨葆胶囊灌胃,连续灌胃42 d.无菌条件下取右侧股骨头,采用光镜观察股骨头病理变化,蛋白免疫印迹(Western Blot)法检测股骨头组织磷酸化细胞外信号调节激酶(p-ERK)1/2、磷酸化c-Jun N-末端激酶(p-JNK)、磷酸化p38(p-p38)蛋白的表达.[结果]造模组家兔股骨空骨陷窝率显著高于空白组(P<0.05),表明造模成功.与模型对照组比较,川骨片治疗组、仙灵骨葆胶囊治疗组股骨头病理改变明显减轻,p-ERK1/2蛋白表达显著升高(P<0.05);p-JNK、p-p38蛋白表达显著下降(P<0.05),且2个治疗组间比较,差异无统计学意义(P>0.05).[结论]川骨片可能是通过上调股骨头p-ERK1/2的表达,下调p-JNK、p-p38的表达,促进激素性股骨头缺血性坏死家兔新骨的形成,抑制骨的破坏,促进骨坏死的修复.
Objective To observe the mechanism of Chuangu Tablets for the treatment of steroid-induced avascular necrosis of femoral head in rabbits.Methods A total of 56 Japanese male rabbits were randomly divided into blank group(16 rabbits)and modeling group(40 rabbits).A rabbit model of steroid-induced avascular necrosis of femoral head was established by HE Xi-Jing method.After 6 weeks,4 rabbits from the blank group and 4 rabbits from the modeling group were executed to judge whether the model was successful and then the rate of empty bone lacunae was calculated.After successful modeling,the modeling group was randomly divided into model control group,Chuangu Tablets treatment group and Xianling Gubao Capsules treatment group,12 rats in each group.The blank group was given no special treatment,and the other 3 groups were correspondingly respectively given intragastric administration of distilled water,Chuangu Tablets and Xianling Gubao Capsules for 42 continuous days.Then the right femoral head was taken out under aseptic condition,and the pathological feature of femoral head tissue was observed under light microscope.The expression levels of phosphorylated extracellular signal-regulated kinase(p-ERK)1/2,phosphorylated c-Jun N-terminal kinase(p-JNK)and phosphorylated p38(p-p38)protein in femoral head tissue were detected by Western blotting method.Results The empty lacunae rate of femur in the modeling group was significantly higher than that in the blank group(P<0.05),showing the modeling was successful.Compared with the model control group,the pathological features of femoral head tissue were relieved in Chuangu Tablets treatment group and Xianling Gubao Capsules treatment group,and the protein expression level of p-ERK1/2 was significantly higher(P<0.05),the protein expression levels of p-JNK and p-p38 were significantly lower(P<0.05),there being no significant difference between the two treatment groups.Conclusion Chuangu Tablets can inhibit bone destruction and promote the repair in rabbit steroid-induced avascular necrosis of femoroal head possibly through up-regulating the expression of p-ERK1/2 and down-regulating the expression of p-JNK and p-p38 in the femoral head thus to promote the formation of new bone.
