摘要
为建立高效的甘薯叶绿体分离方法,比较了2种叶绿体分离方法,即高盐-低pH法和Percoll密度梯度法.研究显示,2种方法都能分离得到完整、纯净的叶绿体.再用CTAB法可从纯叶绿体中提取DNA,用2种方法分离的叶绿体DNA经分光光度计检测,高盐-低pH法和Percoll密度梯度法A260/A280数值分别为1.81和1.89,高盐-低pH法分离得到的叶绿体DNA产率更高,鲜质量分数为270 ng/g;Percoll密度梯度法分离得到的叶绿体DNA产率较低,鲜质量分数为90 ng/g.该研究为深入探究甘薯叶绿体基因组结构、功能等提供基础.
To find the efficient separation method for sweetpotato chloroplast, the high-salt low-pH method and Percoll density gradient method were compared in this study. The results showed that the unbroken and pure chloroplasts could be obtained by both methods, and then the DNA could be extracted from the pure chloroplast by CTAB method.The A260/A280 values of chloroplast DNAs extracted by high-salt low-pH method and Percoll density gradient method were 1.81 and 1.89 respectively.The fresh weight content(270 ng/g) of chloroplast DNA obtained by high-salt low-pH method was higher than that(90 ng/g) obtained by Percoll density gradient method. These results laid a foundation for further on the structure and function study of sweetpotato chloroplast genome.
