摘要
目的探讨三七皂苷R1(NR1)对乳腺癌组织血管生成的影响。方法体外研究采用MTT实验检测NR1对乳腺癌细胞生长的影响;采用ELISA试剂盒检测血管紧张素Ⅱ(AngⅡ)及NR1对乳腺癌细胞培养上清液中血管内皮生长因子(VEGF)浓度的影响;采用Western blotting技术检测AngⅡ和NR1对乳腺癌细胞CD34蛋白表达的影响。体内实验利用裸鼠移植瘤模型检测NR1对乳腺癌移植瘤生长的影响;并利用免疫组化技术检测NR1对移植瘤组织中CD34、VEGF蛋白表达的影响。结果AngⅡ剂量依赖性地上调乳腺癌细胞CD34蛋白的表达以及乳腺癌细胞培养上清液中VEGF的浓度,而NR1可以剂量依赖性地逆转该过程。体内实验结果显示,NR1可以抑制乳腺癌移植瘤小鼠肿瘤的生长,下调荷瘤裸鼠肿瘤组织中CD34、VEGF的表达。结论NR1可能通过下调乳腺癌细胞CD34蛋白的表达及VEGF的产生从而抑制乳腺癌血管的生成,该研究为乳腺癌治疗药物的研发提供了一定的理论依据。
Objective To investigate the effect of notoginsenoside R1(NR1)on angiogenesis in breast cancer tissue.Methods As for in vitro experiments,MTT assay was used to analyze the effect of NR1 on the growth of breast cancer cells;ELISA was used to investigate the effect of angiotensinⅡ(AngⅡ)and NR1 on the concentration of vascular endothelial growth factor(VEGF)in the culture supernatant of breast cancer cells;Western blotting was used to analyze the effect of AngⅡand NR1 on the protein expression of CD34 in breast cancer cells.As for in vivo experiments,the xenograft tumor model in nude mice was used to observe the effect of NR1 on the growth of breast cancer xenograft tumor,and immunohistochemistry was used to investigate the effect of NR1 on the protein expression of CD34 and VEGF in xenograft tumor.Results AngⅡupregulated the protein expression of CD34 in breast cancer cells and the concentration of VEGF in the culture supernatant of breast cancer cells in a dose-dependent manner,while NR1 reversed this process in a dose-dependent manner.In vivo experiments showed that NR1 inhibited the growth of tumor in mice with breast cancer xenograft tumor and downregulated the expression of CD34 and VEGF in the tumor tissue of nude mice.Conclusion NR1 can inhibit the angiogenesis of breast cancer by downregulating the protein expression of CD34 and the production of VEGF in breast cancer cells,which provides a theoretical basis for the research and development of therapeutic drugs for breast cancer.
