5-羟色胺前体物对围产期母羊肝脏糖异生关键基因mRNA和蛋白质表达量的影响 被引量:1

Effects of Serotonin Precursor on mRNA and Protein Expressions of Liver Gluconeogenesis Key Genes of Perinatal Ewes

摘要 本试验旨在研究5-羟色胺(5-HT)前体物对围产期母羊肝脏糖异生关键基因mRNA和蛋白质表达量的影响。试验选择体况良好、体重为(64.52±4.11)kg、处于同一围产期的30只巴美肉羊母羊,按体重随机分为3组,分别为对照组(灌注生理盐水)、5-羟基色胺酸(5-HTP)组(灌注5-HTP)和色氨酸(Trp)组(灌注Trp),每组10只。在母羊围产期产前第7天至产后当天(第0天)进行颈静脉灌注,5-HTP和Trp的灌注剂量均为0.178 mg/kg BW,浓度为0.1 mg/mL;对照组灌注同等剂量的生理盐水。试验期为产前第7天至产后第30天。结果表明:1)5-HTP组和Trp组的肝脏磷酸烯醇丙酮酸羧激酶(PEPCK)、葡萄糖-6-磷酸酶(G6P)和丙酮酸羧化酶(PC)的mRNA表达量均显著高于对照组(P<0.05)。2)5-HTP组和Trp组的肝脏PEPCK、PC和G6P的蛋白质表达量均显著高于对照组(P<0.05),Trp组的肝脏PEPCK的蛋白质表达量显著低于5-HTP组(P<0.05),Trp组的肝脏G6P的蛋白质表达量显著高于5-HTP组(P<0.05)。由此可见,5-HT前体物不仅在转录水平,也在翻译水平起到促进肝脏糖异生的作用。 This study was conducted to investigate the effects of serotonin(5-HT)precursor on mRNA and protein expressions of liver gluconeogenesis key genes of perinatal ewes.Thirty good body condition Bamei ewes with a body weight of(64.52±4.11)kg in the same perinatal period were randomly divided into 3 groups according to their body weight,which were control group,5-hydroxy-tryptophan(5-HTP)group(perfused 5-HTP)and tryptophan(Trp)group(perfused Trp),respectively,each group contained 10 ewes.Ewes jugular vein perfusion was conducted from prenatal day 7 to postpartum day 0,the perfusion dose of 5-HTP and Trp was 0.178 mg/kg BW,and the concentration was 0.1 mg/mL;the control group was perfused with the same dose of saline.The experiment lasted form prenatal day 7 to postpartum day 30.The results showed as follows:1)the mRNA expressions of phosphoenolpyruvate carboxykinase(PEPCK),glucose-6-phosphatase(G6P)and pyruvate carboxylase(PC)in liver of 5-HTP group and Trp group were significantly higher than those of the control group(P<0.05).2)The protein expressions of PEPCK,G6P and PC in liver of 5-HTP group and Trp group were significantly higher than those of the control group(P<0.05),the liver PEPCK protein expression of Trp group was significantly lower than that of 5-HTP group(P<0.05),and the liver G6P protein expression of Trp group was significantly higher than that of 5-HTP group(P<0.05).In conclusion,5-HT precursor plays a role in promoting the liver gluconeogenesis not only at the transcriptional level,but also at the translation level.
