Determination of choline in food for special medical purpose by ultra–high performance liquid chromatography–tandem mass spectrometry

摘要 目的 建立超高液相色谱–串联质谱法(ultra high performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)测定配方食品中胆碱的分析方法 ,为胆碱及其制剂的含量测定提供参考.方法样品中的胆碱经过盐酸水解,提取后,以0.1%甲酸-甲醇作为流动相经kinetex 5u XB-C18柱(4.6 mm×100 mm,5μm)梯度洗脱分离,流速0.45 mL/min,采用UPLC-MS/MS在正离子模式下检测,外标法定量.结果 在优化实验条件下,测得胆碱在浓度为0.1~0.5μg/mL有良好线性关系,线性相关系数为0.9993.方法的回收率为96.1%~103.4%,检出限为2.27 mg/100 g.结论 超高效高效液相色谱–串联质谱法简单快速、灵敏度高、准确性强,适用于特殊医学用途配方食品中胆碱的分析. Objective To establish an analytical method for the determination of choline in formula foods by ultra-high liquid chromatography-tandem mass spectrometry, it provides a reference for the determination of choline and its preparations. Methods The choline in the sample was hydrolyzed and extracted by hydrochloric acid. The separation was performed on the chromatographic column kinetex 5 u XB-C18(4.6 mm×100 mm, 5 μm) with gradient elution using 0.1% formic acid-methanol as mobile phase, the flow rate was 0.45 mL/min. UPLC-MS/MS was used for detection in positive ion mode, and quantitative measurement was conducted by external standard method. Results Under optimized experimental conditions, choline concentration of 0.1–0.5 μg/mL had a good linear relationship, and the linear correlation coefficient was 0.9993. The recovery rate of the method was 96.1%–103.4%, and the detection limit was 2.27 mg/100 g. Conclusion High performance liquid chromatography-tandem mass spectrometry is simple, rapid, sensitive and accurate, suitable for the analysis of choline in food for special medical purpose.
