约氏乳杆菌对LPS诱导的奶牛子宫内膜上皮细胞炎症反应的影响 被引量:4

摘要 子宫内膜炎是影响奶牛生产的重要疾病之一,该病的有效防控至关重要。本试验探讨了约氏乳杆菌对LPS诱导的奶牛子宫内膜上皮细胞炎症反应的预防作用。约氏乳杆菌悬浮至104,105,106 CFU/mL后,与奶牛子宫内膜上皮细胞共同培养3h,添加1mg/L LPS处理3h,检测趋化因子IL-8和促炎因子TNF-αmRNA、蛋白分泌量和NF-κB信号通路关键蛋白表达量。结果表明,LPS处理奶牛子宫内膜上皮细胞3h后,IL-8和TNF-αmRNA和蛋白分泌量均显著上升(P<0.05),104 CFU/mL约氏乳杆菌预处理后IL-8、TNF-α表达和NF-κB信号通路关键蛋白均显著降低(P<0.05),而105或106 CFU/mL约氏乳杆菌预处理并未影响IL-8和TNF-α表达。结果提示,约氏乳杆菌具有预防奶牛子宫内膜炎的作用,且104 CFU/mL表现出良好的抗炎效用。 Endometritis is one of the most important diseases affecting the dairy cow industry,and an effective prevention and control of this disease is essential.The present study aimed to demonstrate the preventive effect of Lactobacillus johnsonii on LPS-induced inflammatory response in the bovine endometrial epithelial cells.After the suspension of Lactobacillus johnsonii pretreatment in 104,105 and 106 CFU/mL,it was co-cultured with bovine endometrial epithelial cells for 3h,and then treated also with 1mg/L LPS for another 3h.The expression of chemokine IL-8,pro-inflammatory factor TNF-αand the key proteins of NF-κB signaling pathway were determined.The results showed that the expression of IL-8and TNF-αmRNA and thier protein were significantly increased after the bovine endometrial epithelial cells were treated with LPS for 3h,most importantly IL-8after 104 CFU/mL Lactobacillus johnsonii pretreatment in comparison to the 105 or 106(P<0.05).In contrast,both TNF-αand protein expression were significantly decreased(P<0.05),while 105 or 106 CFU/mL Lactobacillus johnsonii pretreatment did not affect IL-8and TNF-αexpression(P>0.05).The results suggest that Lactobacillus johnsonii has the effect of preventing endometritis in dairy cows,and Lactobacillus johnsonii pretreatment 104 CFU/mL shows good anti-inflammatory effects.
