摘要
冻精的生产和应用可充分保存和利用优秀种畜的遗传资源并加快品种改良进程.然而,目前绵羊的冻精技术体系还不完善.戴瑞羊(DairyMeade)是国内引进的新型乳用绵羊品种,为建立适用于戴瑞羊的细管冻精生产方案,本研究选用3只18月龄的戴瑞种公羊(N1、N2、N3),在冷冻曲线的-10℃^-100℃危险温区内设置8个降温速率(30℃/min^100℃/min,间隔10℃/min),利用程序化冷冻仪生产细管冻精,并比较了冻后精子的质膜完整率、线粒体膜完整率和IVF后的胚胎卵裂率.结果显示:1、N1羊60℃/min、70℃/min和80℃/min三个降温速率下冻后精子质膜完整率无显著差异,但显著高于其他5个降温速率组(P<0.05);N2与N3羊60℃/min降温速率下的冻后精子质膜完整率与80℃/min组差异不显著,但显著高于其他6个降温速率组(P<0.05);2、三只羊60℃/min降温速率下冻后精子的线粒体膜完整率与70℃/min降温速率组无显著差异,但显著高于其他6个降温速率组(P<0.05);3、60℃/min组冻后精子体外受精后的胚胎卵裂率最高.结果表明,在-10℃^-100℃降温区间内60℃/min的降温速率可获得质量较好的戴瑞羊细管冻精.本研究为戴瑞奶绵羊冻精程序的优化提供了理论依据.
The production and utilization of frozen semen could preserve and make full use of the genetic materials of the elite stud animal,and promote the breed improvement efficiency.However,the current ovine semen frozen system is far behind practical usable.DairyMeade is a specific dairy sheep breed which was newly introduced into China.To establish frozen semen production protocol for DairyMeade,three rams(N1,N2,N3) at the age of 18 months were chosen and their semen were diluted and frozen under 8 different freezing rates(from 30℃/min to 100℃/min,10℃ interval) during the temperature range of-10℃^-100℃ which was recognized as the "dangerous temperature zone",then the plasma membrane integrity,mitochondrial membrane integrity and embryo cleavage rate after IVF of post-thaw sperm were examined and compared.The results were as follows.(1) The post-thaw sperm plasma membrane integrity rates of N1 in 60℃/min,70℃/min and 80℃/min freezing rate groups had no significant differences,but they were significantly higher than the other 5 groups(P<0.05).For N2 and N3,the plasma membrane integrity rate of 60℃/min group had no significant difference with 80℃/min,but was significantly higher than the other 6 groups(P<0.05).(2) The mitochondrial membrane integrity of the post-thaw sperm from at the freezing rate of 60℃/min had no significant difference with 70℃/min group,but was significantly higher than the other 6 groups for each ram(P<0.05).(3) The IVF embryo derived from the post-thaw sperm of 60℃/min group yielded the best cleavage rate.The results indicated that the freezing rate of 60℃/min during the critical freezing stage yielded better post-thaw sperm.This study provided theoretical and practical basis for optimizing the frozen semen protocol for DairyMeade sheep.
