摘要
目的:观察雄黄作用于骨髓增殖性肿瘤HEL细胞的抑制率及对JAK2基因表达的影响。方法:通过体外细胞培养骨髓增殖性肿瘤HEL细胞冻存、复苏、培养,按照不同药物浓度的雄黄液作用于HEL细胞,噻唑蓝法检测HEL细胞抑制率,流式细胞仪检测细胞凋亡,RT-PCR法检测JAK2基因mRNA表达。结果:雄黄可明显抑制HEL细胞的增殖,并且随雄黄剂量的增加,抑制作用增强,JAK2 mRNA的表达减弱,此结果与时间相关。结论:推断雄黄通过作用于JAK2/STAT信号通路,而达到抑制骨髓增殖性肿瘤HEL细胞增殖的作用。
Objective: To observe the effects of Realgar on the inhibitory rate of myeloproliferative neoplasms HEL cells and the expression of JAK2 gene through cell culture in vitro. Methods: Myeloproliferative neoplasms HEL cells were frozen, resuscitated and cultured. The different concentrations of Realgar effects on myeloproliferative neoplasms HEL cells. The inhibition rate of HEL cells was detected by thiazole blue method, apoptosis was detected by flow cytometry, and JAK2 gene expression was detected by RT-PCR. Results: Realgar significantly inhibited the proliferation of HEL cells, and with the increase of Realgar dosage, the inhibition was enhanced, and the expression of JAK2 mRNA was weakened, which was related to time. Conclusion: Realgar can inhibit the proliferation of the myeloproliferative neoplasms HEL cell by acting on JAK2/STAT signaling pathway.
