不同浓度的丙泊酚对人胃癌BGC-823细胞增殖以及相关蛋白表达的影响 被引量:3

摘要 目的评价不同浓度的丙泊酚对胃癌细胞BGC-823细胞增殖周期以及相关蛋白表达的影响。方法将BGC-823细胞用含有10%的胎牛血清、1%青霉素/链霉素的1640培养基置于37℃、5%CO-2培养箱里进行培养,当细胞稳定地进入对数生长期时,使用随机数表法将其分为6组,分别为对照组(C组);脂肪乳组(E组)加入10μg/mL脂肪乳;2μg/mL丙泊酚组(P1组);4μg/mL丙泊酚组(P2组);8μg/mL丙泊酚组(P3组);16μg/mL丙泊酚组(P4组)分别加入2、4、8、16μg/mL丙泊酚。采用蛋白印迹(Western blot)方法测BGC-823细胞中Caspase-9、Caspase-3、BCL-2蛋白的表达情况。结果与对照组(C组)相比较,E、P1、P2组OD值差异无统计学意义(P>0.05);但随着丙泊酚药物浓度的增加,P3、P4组OD值显著降低,差异具有统计学意义(P<0.05)。蛋白印迹结果显示,E、P1、P2、P3组与对照组(C组)相比较,Caspase-9的表达量无显著变化,差异无统计学意义(P>0.05);随着丙泊酚浓度的增加,P4组中Caspase-9的表达显著增加,差异具有统计学意义(P<0.05);E、P1、P2组与对照组(C组)相比较,Caspase-3的表达量无显著变化,差异无统计学意义(P>0.05);随着丙泊酚浓度的增加,P3、P4组中Caspase-3的表达显著增加,差异具有统计学意义(P<0.05);E、P1、P2组与对照组(C组)相比较,BCL 2的表达量无显著变化,差异无统计学意义(P>0.05);随着丙泊酚浓度的增加,P3、P4组中BCL 2的表达显著下降,差异具有统计学意义(P<0.05)。结论丙泊酚可抑制胃癌BGC-823细胞的增殖,其机制与上调caspase 3、caspase 9及降低BCL-2的表达有关。 ObjectiveTo evaluate the effects of different concentrations of propofol on the proliferation cycle and apoptosis of gastric cancer cell line BGC-823.MethodsBGC-823 cells were cultured in 1640 medium containing 10%fetal bovine serum and 1%penicillin/streptomycin at 37℃and 5%CO-2.When the cells steadily entered the logarithmic growth phase,they were divided into six groups:control group(group C),fat emulsion group(group E),10μg/mLL fat emulsion,2μg/mL propofol group(group P1),4μg/mL propofol group(group C),and 4μg/mL propofol group(group E).Propofol group(group P2),8μg/mL propofol group(group P3),16μg/mL propofol group(group P4)were added with 2,4,8 and 16μg/mL propofol respectively.Western blot was used to detect the expression of Caspase-9,Caspase-3 and BCL-2 in BGC-823 cells.ResultsCompared with the control group(group C),the OD values of E,P1 and P2 groups had no significant difference(P>0.05),but with the increase of propofol concentration,the OD values of P3 and P4 groups decreased significantly(P<0.05).Western blotting results showed that the expression of Caspase-9 in E,P1,P2 and P3 groups had no significant change compared with the control group(group C)(P>0.05);with the increase of propofol concentration,the expression of Caspase-9 in P4 group increased significantly(P<0.05);compared with the control group(C),the expression of Caspase-3 in E,P1,P2 group had no significant change trend.There was no significant difference(P>0.05);with the increase of propofol concentration,the expression of Caspase-3 in P3 and P4 groups increased significantly(P<0.05);2)Compared with the control group(C),the expression of BCL-2 in E,P1 and P2 groups had no significant change(P>0.05);with the increase of propofol concentration,the table of BCL-2 in P3 and P4 groups had no significant difference(P>0.05).The apoptotic proteins in P3 and P4 groups increased significantly(P<0.05).ConclusionPropofol can inhibit the proliferation of gastric cancer BGC-823 cells and promote cell apoptosis,which may be related to the up-regulation of caspase 3,caspase 9 and the decrease of BCL-2 expression.
