摘要
目的:了解低血清驯化的悬浮培养型BHK-21细胞的生长特性及致裸鼠成瘤能力.方法:比较研究悬浮型和贴壁型BHK-21细胞的细胞形态、生长动力学和成瘤性.结果:驯化的悬浮培养型和贴壁培养型BHK-21细胞生长状态均良好,最大增殖浓度分别为517×10^4/mL和39.67×10^4/mL,群体倍增时间分别为23.03 h和21.36 h,能显著提高细胞培养密度(P<0.05).背部皮下接种致裸鼠成瘤性实验表明,驯化的悬浮培养型组裸鼠的体重增长率均低于贴壁培养型BHK-21细胞、阳性细胞组(Hela)和阴性细胞组(KMB17)(P>0.05),裸鼠成瘤体积增长率均高于其他三组(P<0.05).结论:驯化的悬浮培养型BHK-21细胞能显著提高细胞密度,其成瘤性方面还需进一步改善.
Objective In order to understand the growth characteristics and the tumorigenic ability to nude mice in suspension cultured BHK-21 cells,which was low serum acclimation in our laboratory,Method The cell morphology,growth kinetics and tumorigenicity of domesticated suspension cultured and adherent cultured BHK-21 cells were studied.Result The results showed that the adherent culture and domesticated suspension culture BHK-21 cells grew well,the maximum proliferation concentrations were 39.67×10^4/mL and 517×10^4/mL,respectively,and the population doubling time was 23.03 h and 21.36 h,respectively.After two subcultures of BHK-21 cells and positive cells(Hela cells)and negative cells(KMB17 cells)were subcutaneously inoculated into nude mice,the growth rate of the nude mice in the domesticated suspension culture group was lower than that of the other three groups.At the same time,the growth rate of the tumor volume of the nude mice was higher than that of the other three groups,and the final tumor volume was also significantly different from the final tumor volume of the nude mice in the positive group and the adherent group(P<0.05).Conclusion Therefore,suspension culture of BHK-21 cells can significantly increase cell density,in terms of suspension culture of BHK-21 cells'tumorigenicity,further improvement is needed.
