摘要
BAK1(BRI1-ASSOCIATED RECEPTORKINASE1)是油菜素内酯信号途径上具有重要功能的植物受体激酶,参与细胞信号转导的许多过程和调控植物生长发育,其在胡杨中的同源基因有2个,分别为PeBAK1;1和PeBAK1;2。本研究克隆了在胡杨中的其中一个同源基因PeBAK1;2,并在烟草中进行了超量表达,通过研究转基因株系对Pst DC3000的抗性表型,以及下游抗病相关基因的表达水平,揭示了PeBAK1;2基因在植物抗丁香假单胞杆菌中的作用和初步的调控机制。抗病表型实验结果显示:与野生型烟草植株相比,PeBAK1;2基因超表达的转基因烟草植株在接种Pst DC3000六天后表现出明显的抗病症状,且转基因叶片中单位面积内的细菌菌落数显著少于野生型;同时,与野生型相比,转基因烟草中与抗病防御相关的Marker基因PR3和PR5的表达量显著升高,且参与植物生长发育与先天防卫反应基因BIR1和BON1的表达量也明显升高;此外,组织表达模式分析实验结果表明,PeBAK1;2基因呈现出组成型的表达模式,但在根和叶片中的表达量较高,在茎中表达量较低。综上所述,我们的研究表明PeBAK1;2基因通过正向调控抗病相关基因和先天防卫反应基因的转录来提高PeBAK1;2转基因烟草对丁香假单胞杆菌的抗性。
BAK1(BRI1-ASSOCIATED RECEPTORKINASE1)is a plant-receptor kinase with an important function in the brassinosteroid signal pathway.It is involved in many processes of cell signal transduction and regulates plant development.PeBAK1;1 and PeBAK1;2 are two homologous genes in Populus euphratica,respectively.In this study,one of the homologous genes in Populus euphratica,PeBAK1;2,was cloned and overexpressed in tobacco.By studying the resistance phenotype of transgenic tobacco lines to Pst DC3000 and the expression level of downstream pathogenesis-resistant genes,we revealed the role of PeBAK1;2 gene in plant resistance to Pst DC3000 and the preliminary regulatory mechanism.The results of disease resistance phenotype show:compared with the wild-type tobacco plants,the transgenic tobacco plants with PeBAK1;2 overexpression showed obvious disease resistance after Pst DC3000 inoculation for 6 days,and the bacterial colony number per unit area in the transgenic leaves was significantly less than that of the wild-type tobacco plants.At the same time,compared with the wild-type,the expressions of pathogenesis-related marker genes PR3 and PR5 in transgenic tobacco were significantly increased,and the expressions of BIR1 and BON1 genes involved in plant development and innate defense responses were also significantly increased.In addition,the results of tissue expression pattern analysis indicated that PeBAK1;2 gene was presented a constitutive expression,but the expression level was higher in roots and leaves,and lower in steams.In conclusion,our study showed that PeBAK1;2 can enhance the resistance of PeBAK1;2 transgenic tobacco to Pst DC3000 by positively regulating the transcription of pathogenesis-related genes and innate defense response genes.
